Abstract:

Technika matrix assisted laser desorption/ionisation (MALDI) time of flight (TOF) mass spectrometry (MS) nachází významné uplatnění v biochemii, lékařství, farmakologii, analytické chemii. Důležitou roli v MALDI TOF MS hraje výběr a použití vhodné marice. Tato diplomová práce se zabývá použitelností různých nanomateriálů a nanočástic jako matric v hmotnostní spektrometrii pro biochemické aplikace, a sledováním jejich vlivu na ionizaci pro stanovení peptidů. Studovány byly možnosti MALDI stanovení peptidů s použitím grafenu, grafen oxidu, nanodiamantů a různých zlatých nanočástic. Efekt nanomateriálů na ionizaci peptidů byl zkoumán ve srovnání s konvenční metodou MALDI. Bylo zjištěno, že intenzity některých peptidových píků (m/z = 750 - 2000) byly 12 × vyšší s použitím polyhedrálních/květinových zlatých nanočástic v kombinaci s nanodiamanty ve srovnání s klasickou matricí (CHCA). Bylo prokázáno zvýšení senzitivity detekce peptidů a potvrzen pozitivní efekt použitých nanomateriálů na desorpci/ionizaci peptidů.

Abstract:

The technique of matrix assisted laser desorption/ionisation (MALDI) time of flight (TOF) mass spectrometry (MS) finds significant use in biochemistry, medicine, pharmacology, analytical chemistry. Selection and application of suitabe matrix is especially important in MALDI TOF MS. The thesis is focused on the possibilities of various nanomaterials and nanoparticles as matrices in mass spectrometry for biological applications, and their influence on the ionisation for the detection of peptides. The possibilities of MALDI for detection of peptides with the use of graphene, graphene oxide, nanodiamonds and different gold nanoparticles have been studied. The effect of these nanomaterials on the ionization of peptides was investigated in comparison with the conventional method of MALDI. It has been proven that the intensities of some of the peptide peaks (m/z = 750 - 2000) were 12 × higher with the usage of polyhedral/flower-like gold nanoparticles in combination with nanodiamonds than with classical matrix (CHCA). Therefore, an increase of the sensitivity of the peptide detection was achieved and it can be concluded that the used nanomaterials have a benefitial effect on the peptide desorption/ionisation.
