Factors playing a role in biological activity of elicitins

Abstract:

Kryptogein a infestin jsou malé proteiny patřící do skupiny elicitinů, sekretovaných oomycetami Phytophthora cryptogea a Phytophthora infestans, vyvolávající obrannou reakci a rezistenci u celé řady rostlinných druhů. V rámci diplomové práce byla exprimována řada mutantních variant infestinu u kterých došlo k záměně aminokyselin na vybraných pozicích a tím ke změně izoel-ektrického bodu samotného protein. Exprese probíhaly v expresním systému kva-sinky Pichia pastoris, která je schopna exprese a správného složení eukaryotních proteinů. Po aplikaci mutantních variant k buněčné suspenzi tabáku se sledovala produkce reaktivních forem kyslíku, respektive míra tvorby peroxidu vodíku, jakož to odpověď rostlinného obranného systému. Odpověď buněk tabáku na mu-tantní varianty infestinu byla porovnávána s odpovědí na samotný elicitin kryp-togein o stejné koncentraci.

Abstract:

Cryptogein and infestin are small proteins characterized as elicitins pro-duced by oomycetes Phytophthora cryptogea and Phytophthora infestans. These proteins induce defensive response and resistance in plants. In this thesis, we cre-ated mutant variants of infestin where mutagenesis was performed at specific locations in a protein sequence leading to changes in its isoelectric point. Proteins were produced in an expression system on P. pastoris, which is capable of eukar-yotic protein expression. Consequently, elicitins were added to the tobacco cell suspension, and reactive oxygen species production was measured as a response of the plant defence system. The reaction of each variant was compared to each other and to the response of the cryptogein at the same concentrations.
