Genetic diversity of harpacticoids (Copepoda: Harpacticoida) of the Bohemian Massif and Western Carpathians with focus on spring fens

Abstract:

Tato diplomová práce se zabývá genetickou diverzitou a příbuzenskými vztahy plazivek z pramenišť Českého Masivu a Západních Karpat metodami fylogenetické analýzy, fylogeografie prostřednictvím haplotypových sítí a analýzou polymorfismu. Rešeršní část se zabývá charakteristikami plazivek, včetně jejich schopnosti se šířit a jejich diverzitě, vlivu glaciace na rozšíření druhů s užším zaměřením na studované území a molekulárním analýzám, především používaným markerům. V rámci praktické části byl modifikován protokol pro extrakci DNA, PCR a purifikaci. Dále bylo získáno 81 mitochondriálních a 39 jaderných sekvencí markerů COI a 28S 7 druhů z 12 lokalit: Attheyella crassa, A. wierzejskii, Bryocamptus echinatus, B. minutus, B. spinolus, B. zschokkei a Canthocamptus staphylinus. Na jejich základě a na dodatečných sekvencích byla odhalena vysoká míra vnitrodruhové divergence u většiny druhů, potenciálně indikující geografickou izolaci či kryptické druhy. U druhu B. spinulosus byla zjištěna přítomnost 2 linií, potenciálně geograficky odlišených.

Abstract:

This diploma thesis deals with genetic diversity and evolutionary relationships of harpacticoids from spring fens of the Bohemian Massif and the Western Carpathians using phylogenetic analysis, phylogeography with the use of haplotype networks and DNA polymorphism analysis. The first part of the thesis summarises characteristics of harpacticoids including their dispersal ability and diversity, the impact of glaciations on species’ ranges with focus on the study area and also concerns the topic of molecular analyses, primarily commonly used markers. In the second part of the thesis, protocols for extraction of DNA, PCR and purification are described. Based on these protocols we obtained 81 mitochondrial and 39 nuclear sequences corresponding to markers COI and 28S, respectively, of 7 harpacticoid species from 12 sites: Attheyella crassa, A. wierzejskii, Bryocamptus echinatus, B. minutus, B. spinolus, B. zschokkei and Canthocamptus staphylinus. Using our and additional sequences we detected high rates of sequence divergence in most studied species which may indicate geographic isolation or presence of cryptic species. In B. spinulosus we discovered 2 lineages which may be geographically distinct.
