Immunosensors for chemical and biological warfare agents detection

Abstract:

Hlavní cíl a náplň této práce byly biosenzory využívající protilátky – imunosenzory – pro detekci biologických agens. Především dva typy biosenzorů byly zvažovány: amperometrické a piezoelektrické vykazující některé výhody (a samozřejmě i nevýhody). Běžně dostupný 10 MHz piezoelektrický QCM a sítotiskové amperometrické elektrody byly použity jako zástupci fyzikálně-chemických převodníků. Práce s biologickými agens je ztížena některými legislativními překážkami. Naše instituce, Centrum biologické ochrany Těchonín, má akreditaci pro zacházení s kmenem Francisella tularensis LVS, proto byla tato bakterie použita jako reprezentativní zástupce. Nakládání s nepatogenními mikroorganismy jako je Bacillus subtilis a Escherichia coli není regulováno, proto byly tyto mikroorganismy v rámci některých pokusů použity. Některé přístupy pro řešení této práce byly zvažovány. Jako jedna z možností bylo měření celých bakteriálních buněk. Tato cesta byla prakticky vyzkoušena použitím bakterií F. tularensis LVS a E. coli. Jinou možností použití imunosenzoru je serologická diagnóza probíhající tularemické infekce v modelu myši BALB/c. Serologická diagnóza byla aplikována za použití jak piezoelektrického tak i amperometrického převodníku. Problematika serologické diagnózy je těsně spojena s charakterizací protilátek. Proto byl piezoelektrický biosenzor využit i pro bližší charakterizaci monoklonálních protilátek tvořených několika hybridomovými buněčnými liniemi. Primárním cílem práce byla příprava biosenzoru, nicméně i jiné metody byly použity při řešení této problematiky. Některé z těchto metod byly dále rozvíjeny ve formě samostatných pokusů. Zvláště pak ELISA (enzyme-linked immunosorbent assay) a pevná fázová extrakce (SPE) byly vybrány pro tyto účely. Výzkumná práce byla nedávno zahájena i v problematice toxických sloučenin, ačkoliv to nebyl původní cíl v rámci zvoleného tématu.

Abstract:

Biosensors utilizing antibodies – immunosensors – are the main aim of this thesis. Especially two types of immunosensors were employed in the presented thesis: amperometric and piezoelectric ones. Each of these biosensors possess some advantages and of course some disadvantages, too. The most common 10 MHz piezoelectric quartz crystal microbalance and screen-printed amperometric electrodes were used as representatives of the physico-chemical transducer and both proved good reliability during experiments towards biosensor-based biological warfare agent detection. Significant legislative obstacles exist for experiments with potential biological warfare agents. Our institution, the Centrum of Biological Defense in Těchonín, has accreditation for experimental work with Francisella tularensis LVS so this bacterium was used as representative and model biological agent. Non-pathogenic microorganisms such as Bacillus subtilis and Escherichia coli can be employed without any regulative obstacles and these species were utilized accordingly. Several approaches were considered for this thesis fulfilment. The development of a method convenient for detection of whole bacterial cells was chosen as one possibility. This detection option was employed for F. tularensis LVS and E. coli assays. In another way, immusensors were developed for serological diagnosis of already manifested tularemic disease in mice BALB/c as model organism. Serological diagnosis was realized not only with the simple piezoelectrical transducer but also with the amperometric one. The field of serological diagnosis is tightly linked with characterization of antibodies, therefore the presented piezoelectric biosensor was furthermore progressively employed for characterization of monoclonal antibodies selected during hybridoma cell lines screening. The primary aim was preparation of biosensors; nevertheless some other methods were employed, too. Some of these methods were finalized into separate manuscripts. Especially the enzyme-linked immunosorbent assay (ELISA) and simple chromatography technique - solid phase extraction (SPE) were selected for this purposes. Recently, research work has been started in the area of chemical toxic compounds even though this was not the primary goal.
