Selected aspects of centrosome biology in tumor cells

Abstract:

Centrosom je organela, která má u živočišných buněk funkci tzv. mikrotubuly organizujícího centra a je tedy klíčová pro správný průběh základních buněčných dějů. Pro karyokinezi je důležitá především bipolarita mitotického vřeténka, které pak zajišťuje bezchybný rozestup sesterských chromatid, resp. celých chromosomů do nově vzniklých dceřiných buněk. Správný počet centrosomů v buňce je proto zásadní pro předcházení chybám během dělení jádra. První část práce je zaměřena na expresi dvou N-terminálních isoforem proteinu p73 – TAp73 a ΔNp73 – ve vztahu k počtu centrosomů v buňkách nádorových linií neurogenního původu. Analýzou endogenní exprese obou isoforem v buňkách s abnormálním počtem centrosomů byla zjištěna spojitost mezi zvýšenou expresí isoformy ΔNp73 a výskytem amplifikace centrosomů. Toto pozorování bylo dále ověřováno na buněčných liniích, u kterých byly jednotlivé isoformy buď overexprimovány nebo knockoutovány. Jednoznačný vztah mezi zvýšenou nebo sníženou expresí těchto isoforem proteinu p73 a abnormálním počtem centrosomů v buňkách však nebyl potvrzen. Druhá část práce vychází z našeho originálního pozorování cytoplazmatického fluorescenčního signálu pro transkripční faktor NANOG v buněčných liniích derivovaných z rabdomyosarkomu. Pomocí imunofluorescenčního značení se potvrdila lokalizace proteinu NANOG v distální oblasti mateřské centrioly. Vzhledem k tomu, že se jednalo o zcela neobvyklou a dosud detailně nepopsanou lokalizaci, byl největší důraz kladen na ověření specificity signálu proteinu NANOG i samotných protilátek. Dále bylo prokázáno, že signál pro protein NANOG se nachází pouze na jednom z obou centrosomých párů a jeho přítomnost je závislá na fázi buněčného cyklu, a tím i na průběhu zrání dceřiných centriol v centrioly mateřské. Vzhledem ke zjištění, že protein NANOG lze detekovat na centrosomech v různých typech nádorových i nenádorových buněk, jsou tyto výsledky zcela originální a poprvé poukazují na možnou funkci tohoto proteinu v biologii centrosomu.

Abstract:

The centrosome is an organelle that serves as a microtubule-organizing center of the animal cell and thus is crucial for the correct sequence of basic cellular processes. Primarily, the bipolarity of the mitotic spindle is critical for accurate segregation of sister chromatids, resp. chromosomes into daughter cells during karyokinesis. Thus, numerical centrosome homeostasis is essential for the prevention of aberrant nuclear division. The first part of this thesis is focused on the relationship between the expression of two N-terminal isoforms of the p73 protein – TAp73 and ΔNp73 – and the number of centrosomes in cells from tumor cell lines of neurogenic origin. Analysis of the endogenous expression of p73 isoforms in cells with an abnormal number of centrosomes showed an association between increased expression of the ∆Np73 isoform and centrosome amplification. This observation was further verified in cell lines with TAp73 or ∆Np73 overexpression or knockout. However, an evident relationship between increased or decreased expression of these p73 isoforms and an abnormal number of centrosomes in cells was not confirmed. The second part of this thesis originates from our original observation of the cytoplasmic fluorescence signal of the NANOG transcriptional factor in rhabdomyosarcoma cell lines. Localization of NANOG protein in the distal region of the mother centriole was confirmed using immunofluorescence staining. Since this was a completely unusual and as yet undescribed localization of NANOG, great emphasis was placed on verifying the specificity of the signals of the NANOG protein and the antibodies themselves. Furthermore, it was confirmed that the signal of the NANOG protein was unevenly distributed around the centrosome pair, and its presence was markedly associated with the cell cycle phase and thus also with the process of centriole maturation. Along with the finding that the centrosomal localization of NANOG was detected in various tumor and non-tumor cell types, these results are original and provide the first evidence suggesting the common centrosome-specific role of the NANOG protein.
