Catalase in monoxenous trypanosomatids

Abstract:

In my work, I deal with the differential gene expression of the catalase, an enzyme in the Leptomonas pyrrhocoris parasite in various parts of the host´s organism Pyrrhocoris apterus. The haemocoel (haemolymph) and the host´s digestive tract were selected as suitable parts of the host body. To evaluate the degree of gene expression, I used the quantitative PCR and the small ribosomal subunit gene (18S) was used as a reference. As part of my research, I also collected 100 Pyrrhocoris apterus individuals from the environment where the autopsy was done, microscopic preparations were prepared, samples were taken and qPCR processed. The results were processed by the relative quantification method and plotted graphically. In the course of the research, the prevalence of infections was determined and a standard PCR was performed to assess the quality of the material obtained and the function of the primers.

Abstract:

Ve své práci se zabývám diferenciální genovou expresí enzymu katalázy v parazitovi Leptomonas pyrrhocoris v různých částech hostitelského organismu Pyrrhocoris apterus (Ruměnice pospolné). Jako vhodné části těla ruměnice byl pro tyto účely zvolen hoemocoel (hemolymfa) a trávicí trakt hostitele. K posouzení míry genové exprese jsem využila metodu kvantitativní PCR a jako referenční byl využit gen pro malou ribozomální podjednotku (18S). V rámci mého výzkumu jsem také nasbírala 100 ruměnic pospolných (Pyrrhocoris apterus) z prostředí, u kterých byla provedena pitva,připraveny mikroskopické preparáty, odebrány vzorky a po zpracování provedena samotná qPCR. Výsledky byly zpracovány metodou relativní kvantifikace a graficky porovnány. V průběhu výzkumu byla také stanovena prevalence infekcí a provedena standartní PCR pro posouzení kvality získaného materiálu a funkce primerů.
