Bacteriome and mycobiome analysis from airway and blood of patients with inhalation injury

Abstract:

Inhalační trauma představuje akutní postižení dýchacích cest způsobené inhalací zplodin hoření nebo páry, toxických a dráždivých látek. Mezi nejčastější sekundární komplikace po inhalaci zplodin patří infekce dýchacích cest. Základním předpokladem pro úspěšnou terapii popálených pacientů s infekčními komplikacemi je precizní mikrobiologická surveillance v čase a vhodná antimikrobiální terapie. Cílem této práce bylo zpracovat literární rešerši k problematice mikrobioty (bakterií a kvasinek) u pacientů s inhalačním traumatem a podílet se na zavedení izolační metody pro následnou sekvenační analýzu bakteriomu a mykobiomu ve vzorcích od těchto pacientů. Byla zoptimalizovaná metodika izolace DNA, polymerázové řetězové rekace a poolování vzorků. Výsledný pool byl předán k dalším krokům. Zavedená metodika bude v navazující studii využita při sledování dynamických změn mikrobioty dýchacích cest u pacientů s inhalačním traumatem.

Abstract:

Inhalation trauma is an acute injury of the respiratory system caused by the inhalation of combustion products or vapors, toxic and irritating substances. The most common secondary complications after inhalation of combustion products include respiratory infection. The basis for successful therapy of burn patients with infectious complications is precise microbiological surveillance in time and suited antimicrobial therapy. The main aims of this thesis were to conduct a research about the issue of microbiota (bacteria and yeast) in patients with inhalation trauma and to participate on introduction of an isolation method for sequencing analysis of bacteriome and mycobiome in samples of these patients. The methodology of DNA isolation, polymerase chain reaction and pooling of samples was optimized. The resulting pool was passed onto the next steps. The established methodology will be used in the following study to monitor dynamic changes of airway microbiota in patients with inhalation trauma.
