Analysis of Eudiplozoon nipponicum (Monogenea) Genome

Abstract:

Eudiplozoon nipponicum je parazitický červ třídy Monogenea, podtřídy Polyopisthocotylea. Krev sající druhy této skupiny, převážně ektoparazité ryb, byly zkoumány především prostřednictvím morfologických a fylogenetických znaků. Tato diplomová práce se zabývá bioinformatickou analýzou a vyhodnocením sekvenačních dat získaných v rámci realizovaného projektu. Z těchto dat se podařilo složit mitochondriální genom tohoto parazita, který byl následně oanotován. Všechny mitochondriální geny typické pro parasitické platyhelmity byly identifikovány a následně použity v rámci fylogenetické analýzy. Evoluční vztahy jak platyhelmintů, tak i třídy Monogenea jsou stále nejasné. Výsledky naší fylogenetické analýzy se příklání k parafylytyckému vztahu třídy Monogenea, což je v rozporu se současnou klasifikací založenou na morfologických i molekulárních datech.

Abstract:

Eudiplozoon nipponicum is a parasitic worm of class Monogenea, subclass Polyopisthocotylea. Blood-sucking species of this class, predominantly fish ectoparasites, have been studied mainly in the context of morphological and phylogenetical characteristics. This diploma thesis is focused on a bioinformatic analysis and evalution of sequencing data gained in the frame of running grant project. The mitochondrial genome of this parasite was assembled from these sequencing data, and subsequently annotated. All the mitochondrial genes characteristic for the parasitic flatworms were identified and subsequently used in the phylogenetic analysis. The evolution relationships of both Platyhelminthes and Monogeneans are still not clear. The results of our phylogenetic analysis suggest the paraphyletic relationship of the Monogenea class, which is in the conflict with the current classification based on both morphological and molecular data.
