Characterization of new organometallic compounds with potential anticancer effects

Abstract:

Cílem biomedicínského výzkumu orientovaného na léčbu nádorových onemocnění je vývoj léčiv vysoce selektivních pro rakovinné buňky s minimálním poškozením zdravých tkání. Stejným směrem je zaměřena i tato dizertační práce cílená na studium organokovových sloučenin, které by mohly významně rozšířit portfolio současných chemoterapeutik. Byly testovány sloučeniny obsahující především železo, ruthenium, ale i další prvky včetně cínu. Poslední jmenovaný byl začleněn do organokovové sloučeniny v kombinaci s nanomateriálem na bázi oxidu křemičitého. Ferroceny jsou z hlediska biologické aktivity považovány za cytotoxické sloučeniny, ale rovněž sloučeniny biokompatibilní, díky obsahu běžně se vyskytujícího železa potřebného ve všech buňkách. Transferinové receptory coby přirozené přenašeče železa do buněk, byly potvrzeny jako klíčové mediátory intracelulární akumulace testovaných ferrocenů. Jako hlavní mechanismus účinku byla detekována indukce oxidativního stresu a poškození mitochondriálních membrán následované spouštěním apoptotických procesů buněčné smrti a v menší míře též autofagie. Podařilo se potvrdit i radiosenzitizující vlastnosti a synergický účinek ferrocenů v kombinaci s cílenou radioterapií. Tato kombinovaná léčba tlumila indukci autofagie a vedla k významnému zvýšení protinádorové účinnosti. V případě nově syntetizovaných ruthenocenů jsme se zaměřili zejména na jejich schopnost tlumit metastazování, které je jedním z nejzávažnějších projevů onkologického onemocnění. Testované rutheniové sloučeniny byly z hlediska působení na nádorové buňky netoxické, ale prokázaly jasnou účinnost ve snižování jejich invazivity a migrace. Tyto sloučeniny ruthenia by mohly v budoucnu působit jako migrastatická chemoterapeutika vhodná pro kombinovanou léčbu metastatických nádorů. Rovněž se nám povedlo navázat spolupráci zaměřenou na výzkum mezoporézních nanočástic oxidu křemičitého v kombinaci s dialkoxid diorganocínem. Hlavním cílem výzkumu nanočástic nesoucích tuto organokovovou sloučeninu byl jejich transport přes membránu díky přímému cílení na folátové receptory. Ty se vyskytují ve vyšším množství u některých typů nádorů. Fragmenty kyseliny folátové významně zvyšovaly intracelulární akumulaci a obohacení dialkoxid diorganocínem zesilovalo cytotoxicitu takto připravených nanočástic. Mechanismus účinku spočíval ve spouštění apoptózy přes indukci oxidativního stresu buněk. Náš výzkum zaměřený na různé typy organokovových sloučenin prokázal jejich potenciál jako možné alternativy současně používaných chemoterapeutik, přičemž rozdílné vlastnosti jednotlivých typů sloučenin jsou příslibem do budoucna pro jejich aplikovatelnost v léčbě různých druhů nádorových onemocnění.

Abstract:

The goal of biomedical research focused on the treatment of cancer is the development of drugs that are highly selective for a cancer cells with minimal damage to healthy tissues. This thesis is focused in the same direction on the study of organometallic compounds which could significantly expand the portfolio of current chemotherapeutic drugs. The compounds containing mostly iron, ruthenium or other including tin were tested. The last named element was incorporated into the organometallic compound with a silica-based nanomaterial. From biological activity point of view the ferrocenes are considered to be cytotoxic but also biocompatible compounds in terms of natural presence of iron essential in all cells. Transferrin receptors as natural transporters of iron into the cells have been confirmed as a key mediators of intracellular accumulation of tested ferrocenes. The main mechanism of action consists in the induction of oxidative stress and damage of mitochondrial membranes followed by triggering apoptotic processes of a cell death, along with the autophagy ongoing in less ratio. The radio sensitizing properties and synergistic effect of ferrocenes in combination with targeted radiotherapy were also confirmed. Such combined therapy was attenuating autophagy induction and resulted in significantly higher anti-tumor efficacy. In case of newly synthesized ruthenocenes, we focused mainly on their ability to suppress metastasis what is one of the most serious manifestations of cancer disease. Tested ruthenium compounds were non-toxic towards tumor cells but revealed clear efficiency in reducing of cell invasiveness and migration. In the future, these ruthenium compounds could act as a migrastatic chemotherapeutics suitable for the combined treatment of metastatic tumors. We also managed to establish cooperation focused on a research of mesoporous silica nanoparticles in combination with dialkoxide diorganotin. The main goal of the research on nanoparticles carrying this organometallic compound was the transport of particles across the membrane for the direct targeting to folate receptors. Their increased amount is occurring in some types of tumors. Folate fragments significantly increased intracellular accumulation and enrichment by dialkoxide diorganotin significantly enhanced the cytotoxicity of such prepared nanoparticles. The mechanism of action consisted in triggering of apoptosis through increased oxidative stress of the cells. Our research on different types of organometallic compounds has presented them as possible alternatives to currently used chemotherapeutics wherein the different properties of the individual types of compounds are a promise for future applicability in the treatment of different types of cancer.
