Neuroimaging biomarkers of Parkinson's disease-like pathology in pharmacological and genetic mouse models

Abstract:

Úvod: Parkinsonova choroba (PD) je progresivní neurodegenerativní onemocnění zapříčiněné mnoha environmentálními a genetickými faktory. Současné možnosti léčby jsou limitované nedostatkem raných biologických ukazatelů umožňujících přesnou diagnostiku v premotorickém stádiu. Zobrazování kurtózy difúze (DKI) je metoda magnetické rezonance (MRI) detekující mikrostrukturální změnu tkání, přičemž využívá vodní difúzi. V této práci byly využity dva myší modely vykazující patologické fenotypy PD za účelem zhodnocení citlivosti DKI pro zachycení patologických změn mozku. Metody: Při práci s přístrojem 9.4 T Bruker BioSpec 94/30 USR jsme použili techniku DKI v několika částech mozku spojených s PD. Využili jsme farmakologický myší model navozený akutní intoxikací metamfetaminem (METH) vykazující silnou neurodegeneraci. Druhým modelem byl geneticky upravený myší kmen, u něhož byla genetickou modifikací navozena akumulace humánního α-synukleinu (TNWT-61). U obou modelů byly provedeny behaviorální testy a DKI v definovaných časových odstupech. Užili jsme dva typy analýzy, a to analýzu oblastí zájmu (region of interest, ROI) a “tract based spatial statistics” (TBSS). Výsledky: Ukazatel průměrné kurtózy (mean kurtosis, MK) byl nejcitlivější hodnotou ve všech experimentech. V METH modelu jsme zaznamenali v časné fázi nižší hodnotu MK v substantia nigra (SN), striatu (STR) a sensorimotorické kůře (CTX), zatímco v pozdní fázi jsme shledali opačné výsledky. TBSS analýza zaznamenala pouze v pozdní fázi narůstající hodnotu MK ve ventrálním jádře lemniscus lateraris a v některých laterárních jádrech thalamu. V TNWT-61 modelu jsme odhalili signifikantní zvýšení MK v mnoha regionech. MK změn přibývalo s věkem myší. Konkrétně MK se zvýšila v STR a thalamu ve 4 časových úsecích (3., 6., 9. a 14. měsíci), v SN v 6., 9. a 14. měsíci, v CTX v 9. a 14. měsíci, a v hippocampu pouze ve 14. měsíci života. V případě TBSS byly zjištěny zněny v 6., 9., a 14. měsíci v několika částech mozku a naznačovaly spojení s nárůstem α-synukleinu. Zdá se tedy, že patologické změny začínají v šedé hmotě a později postupují do bílé hmoty mozkové (WM). Závěr: Výsledky naznačují, že DKI podává unikátní informaci v oblasti nálezu patologických změn, které jsou základem výzkumu symptomatologie podobné PD u obou typů myších modelů. Z tohoto důvodu může DKI sloužit jako diagnostický biomarker v brzkém i pozdním stádiu nemoci s translačním potencionálem a významnou možností zlepšit klinickou diagnostiku PD.

Abstract:

Background: Parkinson’s disease (PD) is a progressive neurodegenerative disorder caused by multiple environmental and genetic factors. Current treatment options are limited also due to lack of an early biomarker allowing accurate diagnosis at premotor stage. Diffusion kurtosis imaging (DKI) is a sensitive magnetic resonance imaging (MRI) method for detection of microstructural tissue changes using water diffusion. Two mouse models reflecting different pathological PD-like phenotypes were used in order to investigate DKI sensitivity to detect pathological brain changes. Methods: Using 9.4 T Bruker BioSpec 94/30USR scanner, we used DKI technique to evaluate water diffusion in several PD related brain regions. We used a pharmacological mouse model based on acute methamphetamine (METH) administration, which features a robust neurodegeneration. The second model was genetic model featuring human α-synuclein accumulation (TNWT-61). A combination of behavioral tests and DKI analysis were applied for both animal models at several time-points. We used two types of analysis – region of interest (ROI) based, and data driven approach “tract based spatial statistics” (TBSS). Results: Mean kurtosis (MK) was the most sensitive readout at all studies. In METH model we detected at 5 days lower MK values in substantia nigra (SN), striatum (STR) and sensorimotor cortex (CTX), while opposite results were observed at 1 month. Furthermore, no changes were detected at 5 days using TBSS and at 1 month, we observed increased kurtosis in ventral nucleus of the lateral lemniscus and some of the lateral thalamic nuclei. In TNWT-61 model we found a significant elevation of MK in the TNWT-61 group compared to that in the wild type (WT) controls, which progressed with age of the mice. Specifically, MK was increased in STR and thalamus at all 4 time-points (3, 6, 9 and 14 months), in SN at 6, 9 and 14 months, in CTX at 9 and 14 months and finally in hippocampus only at 14 months of age. In case of TBSS no changes were detected 3 months, while at 6, 9 and 14 months changes were observed in several brain regions suggesting α-synuclein accumulation-associated changes start in the gray matter and later progress to the white matter (WM). Conclusion: The findings indicate that DKI provides a unique information in detecting the pathological changes that underlie PD-like symptomology in both animal models. Therefore, DKI may serve as diagnostic biomarker at early and late stage with translational potential and relevance for improving the clinical diagnosis of PD.
