Occurrence of Francisella tularensis in haematophagous arthropods

Abstract:

Tularémie je zoonotické onemocnění, jehož význam v posledních letech narůstá. Původcem onemocnění je Francisella tularensis, gramnegativní aerobní bakterie patřící do třídy Gammaproteobacteria a čeledi Francisellaceae. Navzdory skutečnosti, že F. tularensis má dopad na zdraví lidí i zvířat, existuje o aktuálním výskytu tohoto patogenu v hematofágních vektorech pouze velmi málo údajů. Cílem této diplomové práce bylo provést rozsáhlý molekulární skríning různých hematofágních vektorů. Na přítomnost F. tularensis byla testována klíšťata (4348 klíšťat druhu Ixodes ricinus, Dermacentor reticulatus a Haemaphysalis concinna), dále komáři (4100 jedinců Aedes vexans) a muchničky (6900 jedinců Simulium spp.). Kromě již zmíněných hematofágních vektorů bylo testováno 260 vzorků plné krve ulovených zajíců. Pozitivitu vykazovaly pouze 2 vzorky – první ve směsi samic D. reticulatus a druhý vzorek klíštěte D. reticulatus, který byl odebrán ze srnce. Oba pozitivní vzorky byly sekvenovány a byla potvrzena přítomnost F. tularensis subsp. holarctica. Přítomnost F. tularensis v komárech a muchničkách nebyla prokázána. Naše práce zdůrazňuje potřebu komplexní surveillance F. tularensis v hematofágních vektorech, rezervoárech i u lidí, pro dosažení lepší prevence a kontroly dříve opomíjeného onemocnění.

Abstract:

Tularemia is a zoonotic disease with an increasing impact in recent years. The causative agent is Francisella tularensis, a gram-negative aerobic bacterium belonging to the class of Gammaproteobacteria and the family Francisellaceae. Despite its undeniable importance for human and animal health, there are few data on the current distribution of F. tularensis in hematophagous vectors. The aim of this thesis was to perform a mass molecular screening of different hematophagous vectors: ticks (4348 ticks of the species Ixodes ricinus, Dermacentor reticulatus and Haemaphysalis concinna), mosquitoes (4100 specimens of Aedes vexans) and black flies (6900 specimens of Simulium spp.) for the presence of F. tularensis. In addition, we also tested 260 blood samples from hunted hares. Only two positive samples were found in the study (one from a pool of D. reticulatus females) and the second from a D. reticulatus tick collected from roe deer. Both positive samples were sequenced and confirmed the status of F. tularensis subsp. holarctica. In addition, the absence of F. tularensis in mosquitoes and black flies was documented. Our study highlights the need for comprehensive surveillance of F. tularensis in haematophagous vectors, reservoir hosts, and humans to achieve better prevention and control of this previously neglected disease.
