Coevolution and codiversification of the host-parasite system Cichlidae-Monogenea

Abstract:

Ústředním tématem předložené disertační práce je studium evolučních mechanismů ovlivňujících biodiverzitu monogeneí parazitujících u ryb čeledi Cichlidae. Vybraným modelem pro toto studium je hostitelsko-parazitický systém Cichlidae z oblasti západní Afriky a jejich monogenea řádu Dactylogyridea. Hlavním cílem disertační práce je (1) studium biodiverzity parazitů řádu Dactylogyridea třídy Monogenea u hostitelských druhů ryb čeledi Cichlidae v Západní Africe, (2) fylogenetická rekonstrukce parazitů a hostitelů na základě molekulárních dat, (3) studium evolučních mechanismů ovlivňujících diverzifikaci a speciaci parazitů rodu Cichlidogyrus a Scutogyrus na základě kofylogenetických analýz a (4) studium determinantů a procesů souvisejících s hostitelskou specifitou těchto parazitů. Africké ryby čeledi Cichlidae jsou pro svou vysokou morfologickou rozmanitost, která je výsledkem jejich adaptivní radiace a rychlé diverzifikace, předmětem zájmu mnoha studií. Africké cichlidy jsou v současnosti parazitovány pěti rody monogeneí řádu Dactylogyridea: Cichlidogyrus Paperna, 1960, Scutogyrus Pariselle & Euzet, 1995, Onchobdella Paperna, 1968, Enterogyrus Paperna, 1963 a Urogyrus Bilong Bilong, Birgi & Euzet, 1994. Ektoparaziti rodů Cichlidogyrus, Scutogyrus a Onchobdella parazitují na žábrách hostitelských ryb, endoparaziti rodů Enterogyrus a Urogyrus se vyskytují v žaludku nebo močovém měchýři svých hostitelů. Na sedmi lokalitách v Národním parku Niokolo Koba byly na sedmi druzích hostitelských ryb čeledi Cichlidae zaznamenány čtyři rody monogeneí řádu Dactylogyridea. Celkem bylo nalezeno 32 druhů monogeneí, dva druhy byly popsány jako nové pro vědu. Fylogenetická analýza na základě molekulárních dat odhalila polyfyletický původ rodu Cichlidogyrus a monofyletický původ rodu Scutogyrus. Rody Enterogyrus a Onchobdella formovaly monofyletickou skupinu, pozice těchto rodů ve fylogenetickém stromě naznačuje, že endoparazitismus monogeneí pravděpodobně u cichlid není ancestrální. Morfologické znaky příchytného aparátu (haptoru) byly mapovány do fylogenetického stromu parazitů. Získané výsledky naznačují, že příchytný aparát se pravděpodobně vyvíjel od nejjednodušších ke komplexnějším formám. Role kospeciace v tomto hostitelsko-parazitickém systému byla testována pomocí kofylogenetických metod založených na topologii fylogenetických stromů nebo na fylogenetických vzdálenostech. Vzhledem k vysokému počtu detekovaných koevolučních událostí typu intrahostitelská speciace (tj. duplikace parazitů) usuzujeme, že duplikace v kombinaci s přeskokem hostitelů („host switching“) mohou vysvětlit diverzifikaci monogeneí rodů Cichlidogyrus a Scutogyrus parazitujících cichlidy v Západní Africe. Dosažené výsledky dále naznačily, že hostitelská specifita zástupců rodů Cichlidogyrus a Scutogyrus není determinována fylogenezí parazitů, ale může odrážet i jiné než historické vlivy. Výsledky mapování hostitelské specifity do fylogenetického stromu parazitů naznačují, že intermediální specialisti, kteří parazitují na kongenerických hostitelských druzích, reprezentují původní charakter hostitelské specifity zástupců rodů Cichlidogyrus a Scutogyrus. Hypotéza specializace na predikovatelné zdroje a hypotéza ekologické specializace nebyly u parazitů rodů Cichlidogyrus a Scutogyrus potvrzeny. Bylo prokázáno, že fylogeneze cichlid a charakter rodičovské péče o potomstvo mohou představovat významné determinanty hostitelské specifity těchto parazitů.

Abstract:

The central topic of this thesis is the study of evolutionary mechanisms affecting the diversity of monogeneans in the host-parasite system Cichlidae-Monogenena in West Africa. The selected host-parasite model for this study is Cichlidae from West Africa and their oviparous monogeneans (belonging to Dactylogyridae). The main objectives of the thesis were: (1) to investigate biodiversity of dactylogyridean monogenean parasites of Cichlidae in West Africa, (2) to perform phylogenetic reconstruction of parasites and their hosts based on molecular phylogenetic data, (3) to investigate phylogenetic and evolutionary patterns potentially linked to diversification and speciation of cichlid dactylogyridean monogeneans using cophylogenetic analyses and (4) to investigate potential determinants and patterns connected with host specificity of these parasites. African freshwater fish of Cichlidae are due to their high morphological diversity linked to their adaptive radiation and diversification subject of interest for many scientists. African cichlids are parasitized by five genera of monogeneans belonging to the Dactylogyridea: Cichlidogyrus Paperna, 1960, Scutogyrus Pariselle & Euzet, 1995, Onchobdella Paperna, 1968, Enterogyrus Paperna, 1963 and Urogyrus Bilong Bilong, Birgi & Euzet, 1994. Ectoparasitic Cichlidogyrus, Scutogyrus and Onchobdella are located on the gills, endoparasitic Enterogyrus and Urogyrus are located in the stomach and urinary bladder, respectively. Representatives of four dactylogyridean genera of monogeneans were collected from six cichlid species on seven localities in the Niokolo Koba National Park. A total of 32 species of parasites have been identified; two Cichlidogyrus species were described as new species. Phylogenetic analyses based on molecular data supported the polyphyletic origin of Cichlidogyrus and suggested that Scutogyrus is monophyletic. Both Enterogyrus and Onchobdella were found to be monophyletic. The position of Enterogyrus and Onchobdella in the phylogenetic tree suggests that endoparasitism in cichlid monogeneans is not an ancestral feature. Mapping of morphological characters of the attachment apparatus of parasites onto the parasite phylogenetic tree suggests that the attachment apparatus has evolved evolved from a very simple form to a more complex one. Two types of methods of cophylogenetic analyses were used to study coevolution in Cichlidogyrus/Scutogyrus and cihlid fish system: a distance-based method and a tree-based method. The high number of intrahost speciations, i.e. parasite duplications, suggests that duplication in combination with host switching is the main coevolutionary event explaining the diversification of monogeneans of Cichlidogyrus and Scutogyrus genera living on West African cichlids. Our results showed that host specificity of Cichlidogyrus and Scutogyrus species is not linked to parasite phylogeny suggesting that specificity may reflect other than historical constrains. The mapping of host specificity onto the parasite phylogenetic tree suggests that intermediate specialist parasitizing congeneric hosts represents the ancestral state for Cichlidogyrus/Scutogyrus group. Our study does not support the specialization on predictable resources and ecological specialization hypotheses. However, we showed that phylogeny and brooding behaviour of cichlids may play an important role for determining host specificity of Cichlidogyrus/Scutogyrus parasites.
