Inhibition of cholinesterases by specific inhibitors

Abstract:

Diplomová práce byla zaměřena na studium kinetiky neinhibovaných a inhibovaných hydrolýz acetylcholinu pomocí acetylcholinesterázy (ACHE). Stanovení probíhalo in vitro pomocí dvou nezávislých metod - HPLC a pH-statové. Inhibiční rychlost jednotlivých inhibitorů byla posuzována podle hodnoty jejich inhibiční rychlostní konstanty k3. Kritériem pro posuzování anticholinesterázové bylo také stanovení hodnoty IC50. Vhodnost využití daných látek jako inhibitorů inhibujících mozkovou acetylcholinesterázu bylo posuzováno na základě výsledků rozdělovacího koeficientu v soustavě n-oktanol/voda. Díky tomu mohla být posouzena prostupnost přes hematoencefalickou bariéru. Ze studované skupiny inhibitorů se jako nejúčinnější z hlediska rychlosti inhibice a schopnosti průniku hematoencefalickou bariérou jeví inhibitory, jejichž alkylem je butylová skupina.

Abstract:

This thesis was focused on the study of kinetics of non-inhibited and inhibited hydrolysis of acetylcholine by acetylcholinesterases (ACHE). The determination was carried out in vitro using two independent methods - HPLC and pH-stat. The inhibitory rate of inhibitors was assessed by the value of the inhibition rate constant k3. The criteria for assessment of antilcholinesterases activity was also determination of IC50 value. The appropriateness of the use of these substances as inhibitors of brain acetylcholinesterase was assessed on the basis of the results of the partition coefficient in n-octanol/water system. With this it could be considered drug permeability through the blood-brain barrier. From the studied group of inhibitors are most effective in term of speed and ability to penetrate blood-brain barrier inhibitors, whose alkyl is a butyl group.
