Efect of anticancer drugs on intercellular communication, respiratory chain and angiogenesis

Abstract:

Během svého postgraduálního studia jsem publikoval tři výzkumné články a tři review v mezinárodních vědeckých časopisech. Tyto práce se staly podkladem pro tuto disertační práci. Během první části svého postgraduálního studia jsem srovnával dvě platinu obsahující protinádorová léčiva, cisplatinu a LA-12 (PtIV komplex ). Prokázal jsem souvislost mezi LA-12 indukovanou apoptózou a inhibicí mezibuněčné komunikace (GJIC), která je založena na LA-12 indukované aktivaci ERK kinázy a následné inhibici mezibuněčné komunikace. Cisplatina narozdíl od LA-12 mezibuněčnou komunikaci neinhibuje. Aktivace ERKu souvisí naopak s apoptózou. Jedná se o zcela nový náhled na mezibuněčnou komunikaci, což je zajímavé téma pro další zkoumání. Zbylá část mého postgraduálního studia byla zaměřena na negenotoxické a neimunosupresivní protinádorové látky. Předmětem mého bádání se stal vitamín E sukcinát. Prokázal jsem, že vitamín E sukcinát interferuje a částečně inhibuje buněčný dýchací řetězec na komplexu II, čímž způsobuje vznik reaktivních kyslíkových radikálů především v nádorových buňkách. Dále jsem se podílel na výzkumu role vitaminu E sukcinátu v angiogenezi, kde hrají kyslíkové radikály také významnou roli. Vitamín E sukcinát se zdá být slibnou protinádorovou látkou, která by měla brzy vstoupit do klinických zkoušek. Disertační práce je rozdělena do čtyř kapitol. První kapitola předkládá základní teoretické informace ke zkoumané problematice. Druhá kapitola podrobněji popisuje méně obvyklé postupy použité během mého studia. Ve třetí kapitole jsou prezentovány a diskutovány dodatečné výsledky, které fakticky podporují a doplňují zmíněné publikace. Obrázky k prezentovaným a diskutovaným výsledkům tvoří čtvrtou kapitolu. K disertační práci jsou také připojeny již zmíněné publikace, kterých jsem autorem nebo spoluautorem.

Abstract:

This Ph.D. thesis is based on three research articles and three review papers that were published during my Ph.D. study on cancer research. I compared two platin based anticancer drugs cisplatin and LA-12 (PtIV complex) during the first part of my Ph.D. study. I have revealed a link between LA-12 induced apoptosis and inhibition of gap junctional intercellular communication (GJIC). I have proved that MAPK pathway is responsible for the inhibition of intercellular communication after LA-12 treatment but is not involved in the process of initiation of apoptosis. But, on the other hand, MAPK pathway is partly involved in cisplatin induced apoptosis. This is a new perspective of the role of intercellular communication and should be further investigated. Over the rest of my Ph.D. study I have focused on non-genotoxic non-immunosuppressive anticancer drugs specifically Vitamin E succinate (α-TOS). I proved that alfa-TOS interferes with and partly inhibits the cellular respiratory chain at the complex II and therefore causes production of reactive oxygen species (ROS) especially in cancer cells. This is an important discovery because it means that the drug is less toxic to non-cancer cells. This lead me to further related research concerning antiangiogenic features of alfa-TOS that is related to production of ROS. Alfa-TOS also seems to be a promising anticancer agent which can soon enter clinical trials. The thesis is divided into four chapters where the first chapter gives a reader the basic information about the problematics discussed in the publications. The second chapter describes the uncommon methods used during my Ph.D. study and the third chapter called Results and discussion presents more facts and results that did not appear in the publications. The last chapter called Supplementary data contains figures and tables mentioned in the results and discussion chapter. It also includes publications attached to the thesis.
