Genetic factors affecting the frequency of t-haplotypes in the house mouse (Mus musculus)

Abstract:

Cílem této bakalářské práce je formou literární rešerše shrnout současné znalosti o t-haplotypu. Jedná se o sekvenci genů v proximální třetině chromozomu 17 děděných jako celek v důsledku čtyř paracentrických inverzí, které brání rekombinaci. Práce se zabývá unikátními vlastnostmi t-haplotypu, které mu svou kombinací umožňují udržet se v populaci ve stavu balancovaného polymorfismu. Zejména schopností vychýlení poměru přenosu vlastních kopií do další generace, a příčinami tohoto vychýlení. K pochopení jejich vlivů nejdříve představí strukturu samotného komplexu a funkce a interakce vybraných genů na něm se nacházejících, zejména genů Tcr („responder“) a Tcd („distorter“), letálních genů, hlavního histokompatibilního komplexu (MHC) a dominantních mutací T. Práce si klade za cíl popsat vlivy těchto genů na frekvenci výskytu t-haplotypu v populacích.

Abstract:

The goal of this thesis is to summarize current knowledge on the t-haplotype. It is a genomic segment located in the proximal third of chromosome 17 inherited as a whole due to four paracentric inversions which supress recombination. This thesis focuses on unique features of t-haplotype, which in combination give it the ability to maintain balanced polymorphism in populations. One of the best known features is the ability to distort the ratio of transmission to the next generations. This paper also concerns itself with the genetic causes of these features. To understand the final effect these features provide, this work introduces the structure of the t-complex and the fuction and interaction of some genetic loci, particularly the responder and distorter genes, lethal factors, the major histocompatibility complex (MHC) and dominant T mutations. As a result this thesis describes the influence these genes have on the frequency of t-haplotypes in mouse populations.
