Analyses of soil microbial communities during model bioremediation experiment.

Abstract:

Cílem práce bylo hodnocení půdních mikrobních společenstev z jednotlivých variant bioremediačního experimentu zaměřeného na biodegradační schopnosti dvou aktinobakteriálních kmenů. Jednotlivé varianty byly navrženy tak, aby bylo z výsledků možné odhadnout faktory, které mohly ovlivnit výsledek degradace, např. přídavek sorbentů nebo několik forem železa. Analýzou fosfolipidových mastných kyselin bylo zjištěno zvýšení koncentrace PLFA v každé variantě posledního odběru. Kmen Gordonia rubripertincta CWB byl schopen lepšího růstu na ropné zátěži ve srovnání s kmenem Rhodococcus erythropolis S43. Dále pak při přídavku železa byly zaznamenány rozdíly v celkové koncentraci PLFA, ale biomasa celkových bakterií ani aktinomycet nebyly železem ovlivněny. Přídavek sorbentu (aktivní uhlí) neměl vliv na mikrobiální biomasu. Výsledky doplňují předchozí zjištění, že pro biodegradaci vysokých koncentrací uhlovodíků je vhodnější kmen G. rubripertincta CWB. Avšak i varianty s inokulovaným kmenem R. ertythropolis S43 zaznamenaly nárůst mikrobní biomasy téměř u všech sledovaných skupin vyjma aktinomycet, kmen R. erythropolis S43 tedy přispívá k růstu dalších mikroorganismů.

Abstract:

The aim of this work was to assess the soil microbial communities from individual variants of bioremediation experiment, which was focused on the biodegradation abilities of two actinobacterial strains. The individual variants were designed to be able to cover the factors that could influence the result of biodegradation, for example the addition of sorbents or different forms of iron. Concentration of phospholipid fatty acids (PLFA) exhibited an increase in allvariants in the last sampling. The strain of Gordonia rubripertincta CWB was able to grow better on oil contamination compared to the strain Rhodococcus erythropolis S43. When iron was added, differences in the overall concentration of PLFA were noted, but the biomass of the total bacteria and Actinomycetes were not affected by iron. Addition of sorbent (activated carbon) did not affect microbial biomass. The results complement the previous finding that the G. rubripertincta strain CWB is more suitable for biodegradation of high hydrocarbon concentrations. However, variants with the inoculated R. ertythropolis S43 strain recorded an increase in microbial biomass in almost all monitored microbial groups except of Actinomycetes, which shows that R. erythropolis S43 contributes to the growth of other microorganisms.
