An analysis of the interactions of adenovirus vectors and human cytomegalovirus with host cells

Abstract:

V této práci jsme studovali vliv virových infekcí na buněčné imunitní systémy prostřednictvím analýzy změn signálních drah a změn receptorů na buněčných površích infikovaných buněk. Použili jsme dva různé typy adenovirových vektorů pro genovou terapii, vektor RAd592 AdEasy1 (adenovirus 5 ΔE1/E3) a vektor RAd1640 AdZ2 (adenovirus 5 ΔE1/E3/E4 (se zachovaným E4ORF6)). Následně jsme studovali interakce mezi buňkou a vektorem a s pomocí kvantitativní proteomiky jsme analyzovali změny ve fosforylaci a expresi proteinů v infikovaných buňkách. V infikovaných buňkách byly pozorovány významné změny vrámci proteomu a fosfoproteomu přičemž největší rozdíly týkající se imunitní odpovědi byly identifikovány v antigen prezentující dráze a v dráze PI3K / mTOR. Abychom mohli analyzovat i změny v ubikvitinaci a ubiquitin-like posttranslační modifikaci, byla v naší laboratoři zavedena metoda identifikace peptidů s GG-modifikací pomocí hmotnostní spektrometrie. Změny v přítomnosti receptorů na buněčném povrchu jsme studovali v buňkách infikovaných HCMV. Identifikovali jsme protein UL148 jako nový imunomodulátor HCMV. UL148 způsobí stažení LFA3 molekuly, která je součástí imunologické synapse a její zadržení v endoplazmatickém retikulu. Následné imunologické analýzy poukázaly na význam přítomnosti LFA3 na buněčném povrchu v podpoře reakcí NK-buněk a T-buněk na infekci. Snížená prezentace LFA3 tak vede k významnému poklesu eliminace buněk infikovaných HCMV NK-buňkami a T-buňkami.

Abstract:

In this thesis work the impact of virus infections on cell immune systems were studied by analysing for changes in signalling pathways and changes in receptors on the cell surfaces of infected cells. Two different types of adenovirus gene therapy vectors, RAd592 AdEasy1 (adenovirus 5 ΔE1/E3) and RAd1640 AdZ2 (adenovirus 5 Δ E1/E3/E4 (with non-deleted E4ORF6)), were used. Subsequently, we studied cell-vector interactions and analysed changes in protein phosphorylation and expression in infected cells by using quantitative proteomics. Significant changes in proteomes and phosphoproteomes were observed in infected cells. The biggest differences concern immune responses were identified in both the antigen presenting and PI3K/mTOR pathways. In order to be able to analyse for changes in ubiquitination and ubiquitin-like post-translation modification, a method for identification of GG-tagged peptides in our laboratory was established using mass spectrometry. Changes in the presence of cell surface receptors were studied in cells infected by HCMV. The product of gene UL148 was identified as a new immunomodulator of HCMV that is shown to act by sequestration of LFA3, part of the immunological synapse, in the endoplasmic reticulum. Further immunological analyses show the importance of the presence of LFA3 on the cell surface in promoting NK-cell and T-cell responses to infection. A significant decrease in LFA3 presentation leads to a significant decrease in NK-cell and T-cell mediated killing of HCMV infected cells.
