Abstract:
Metodou iTRAQ/2DLC-MS/MS byly studovány proteomové profily buněčných linií H1299 exprimujících geny pro proteiny p53-His175 (nesbalená forma) a p53-His273 (sbalená forma) bez a s inhibitorem Hsp90, jímž byl 17-AAG. Bylo prokázáno, že po působení 17-AAG dochází u obou buněčných linií ke zvýšené expresi genů pro další „heat shock“ proteiny, jejichž úkolem je nahrazení zablokované funkce Hsp90. V případě …more
Abstract:
Proteomic profiles of cell lines transfected by mutant sequences for proteins p53-His175 (unfolded) and p53-His273 (folded) with/without inhibition of Hsp90 by 17-AAG were studied using iTRAQ/2DLC-MS/MS experiment. In both cell lines there was demonstrated that 17-AAG treatment causes enhanced expression of genes of another heat shock proteins that can replace blocked Hsp90 functions. A difference …more
