Immobilization of hyaluronic acid on nanofibrous substrates

Abstract:

Bakalářská práce se zabývá funkcionalizací povrchu křemičitých nanovlákenných substrátů s cílem imobilizace kyseliny hyaluronové. V teoretické části jsou shrnuty poznatky získané rešerší, zaměřenou na metody zpracování a aplikaci kyseliny hyaluronové ve formě nanostruktur v medicíně. V další části rešerše jsou diskutovány metody modifikace kyseliny hyaluronové a možnosti využití anorganických nanovlákenných substrátů v medicíně. Experimentální část se zabývá optimalizací silylační reakce, kdy jako modifikační činidlo je zde využit 3-aminopropyltriethoxysilan (APTES). Byl sledován vliv koncentrace silylačního činidla a čas reakce na výsledný výtěžek aminoskupin (NH2) kovalentně navázaných na povrch křemičitých nanovlákenných substrátů. V závislosti na výtěžnosti silylace za různých reakčních podmínek byl stanoven optimální postup pro povrchovou modifikaci křemičitých nanovláken. Další část zahrnuje imobilizaci kyseliny hyaluronové na předem modifikovaný nanovlákenný substrát, kde byl zkoumán vliv pH a čas reakce na výsledný výtěžek kyseliny hyaluronové.

Abstract:

The bachelor thesis deals with surface functionalization of silica nanofibrous substrates with the goal of surface immobilizationthe of hyaluronic acid. The theoretical part summarizes the knowledge obtained from literature research, focusing on processing methods and applications of hyaluronic acid in the form of nanostructures in medicine. In the next section of theoretical part, there are discussed modifications of hyaluronic acid and the possibility of using inorganic nanofibrous substrates in medicine. The experimental part deals with the optimization of silanization reaction with the use of 3-aminopropyltriethoxysilane as a modifying agent. In addition, the effect of the silanization agent concentration and reaction time on the final yield of covalently bonded amine group, were also observed. Depending on the yield of silanization under the various reaction conditions, the optimal procedure for surface modification of silica nanofibers was determined. In the case of the hyaluronic acid immobilization procedure the effect of the reaction pH and time was studied.
