Methods for histone purification from human tissue cultures

Abstract:

Dynamické posttranslační modifikace (PTM) histonů vyskytující se především na N-terminálních oblastech histonového řetězce jsou důležitou ale ne zcela objasněnou součástí regulace genů, epigenetické dědičnosti, integrity genomu a jiných jaderných mechanismů. Podmínkou pro sledování kombinace četných histonových modifikací vyskytujících se v určitém biologickém stavu buňky je použití izolačních metod, při kterých nedojde ke ztrátě PTM. Tato bakalářská práce se věnuje metodám purifikace histonů z lidských tkáňových kultur. Při nejpoužívanější metodě izolace histonů, kyselé extrakci z chromatinu, může docházet ke ztrátě některých modifikací, např. fosforylací na určitých pozicích. Praktická část se z tohoto důvodu věnuje novému postupu metody s použitím močoviny a soli, při kterém by měly být labilnější modifikace lépe chráněny. Kromě vyhodnocení úspěšnosti izolace a purifikace histonů, byl porovnán fosforylační stav histonů s histonovými extrakty izolovanými metodou kyselé extrakce z chromatinu s cílem zjistit, zda zvýšení acetylačního stavu histonů vede ke změnám na úrovni fosforylací, byl sledován účinek histondeacetylázového inhibitoru entinostatu u buněčné kultury MEC-1.

Abstract:

Dynamic post-translational modifications (PTMs) of the histones occur predominantly at the N-terminal regions of histone chains. They are important but not completely understood components of gene regulation, epigenetic inheritance, genome integrity and other nuclear mechanisms. The requirement for monitoring a combination of numerous modifications of histones occurring at a certain biological state of the cell is using isolation techniques in which loss of PTMs is suppressed. This bachelor thesis deals with methods of purification of histones from human tissue cultures. During the most widely used method, the acid extraction from chromatin, some modifications may be lost, such as phosphorylation at certain positions. For this reason the practical part of the thesis deals with the new salt-urea method, in which labile modifications should be better protected. The success of isolation and purification of histones was evaluated as well as the comparison of histone phosphorylation status between the latter and histone extracts isolated by acid extraction of chromatin was made. In order to determine whether an increase in the acetylation status of histones leads to changes in the level of phosphorylation, effect of histone deacetylase inhibitor entinostat in cell culture MEC-1 was observed.
