The influence of oxidative stress on the respiration chain of the bacteria Paracoccus denitrificans

Abstract:

In this diploma thesis we focused on the influence of an oxidative stress caused by an oxidative stressor methyl viologen (MV) on two bacterial strains of a bacterium Paracoccus denitrificans, a wild type (Pden_1222) and FnrP- (Pden_2921, FnrP deficient mutant). The portfolio of known effects of the methyl viologen on the growth of the bacterium Paracoccus denitrificans, which manifests itself in a slowed down growth during its cultivation, was extended with the knowledge of the influence of these species on a respiration chain and a denitrification process. It was found out that the rate of reduction of the first enzyme in the denitrification pathway, which is called nitrate reductase, (when we used isolated cell membranes), is for both strains, whether MV was used or not, influenced by the enzymes that are situated before the nitrate reductase. 9-fold difference of the activity of the enzyme nitrite reductase was discovered between Pden_1222 without MV and Pden_1222 with MV and 44-fold difference between Pden_2921 without MV and Pden_2921 with MV. By measuring NO-reductase of Pden_1222 we received expected results – the activity of Pden_1222 without MV was about 3,5-fold of the activity of Pden_1222 with MV. Unexpected and partially surprising is the opposite trend of Pden_2921, when the cells cultivated with MV had 4-fold activity of the cells cultivated without MV. On the level of gene expression, the differences between relative amounts of transcripts were lower when using different strains and cultivation conditions. It is important to mention the lower activity of the terminal oxidases ba3 and aa3 of the cells cultivated with 30 μM MV in contrast to the control cells (for both tribes Pden_1222 and Pden_2921).

Abstract:

V tejto diplomovej práci sa zaoberáme vplyvom oxidačného stresu vyvolaného oxidačným stresorom metylviologénom (MV) na dva kmene baktérie Paracoccus denitrificans, divoký kmeň (Pden_1222) a FnrP- kmeň (Pden_2921, kmeň deficientný v proteíne FnrP). Portfólio známych účinkov metylviologénu na rast baktérie Paracoccus denitrificans, ktoré sa prejavuje v jej spomalenom raste počas kultivácie, bolo rozšírené o znalosti vplyvu tejto látky na respiračný reťazec a denitrifikačnú dráhu. Bolo zistené, že rýchlosť prvého enzýmu denitrifikačnej dráhy, nitrát reduktázy, (pri použití izolovaných bakteriálnych membrán), je pre oba kmene buniek bez ohľadu na to či boli pestované s MV alebo bez neho, závislá na enzýmoch zaradených v respiračnom reťazci pred nitrát reduktázou. 9-násobný rozdiel aktivity enzýmu nitrit reduktasy bol zistený medzi Pden_1222 bez MV a Pden_1222 s MV a 44-násobný rozdiel medzi Pden_2921 bez MV a Pden_2921 s MV. Pri meraní aktivity NO-reduktázy buniek Pden_1222 pestovaných bez MV mali očakávane asi 3,5krát vyššiu enzýmovú aktivitu než Pden_1222 pestované s MV. Nečakaný a do istej miery prekvapujúci bol však opačný trend pre kmeň Pden_2921, kedy bunky pestované s MV oproti kontrole vykazovali 4-krát vyššiu enzýmovú aktivitu. Na úrovni génovej expresie boli rozdiely medzi relatívnym množstvom transkriptov denitrifikačných enzýmov a terminálnych oxidáz za rôznych kmeňových a kultivačných podmienok o dosť nižšie, za dôležitú zmienku stojí všeobecný trend zníženia génovej expresie pri terminálnych oxidázach ba3 a aa3 pre bunky pestované s 30 μM MV oproti kontrole (týka sa aj kmeňa Pden_1222 aj Pden_2921).
