Preparation of thermostable lytic enzyme designed by in-silico methods

Abstract:

V globálním měřítku komplikuje léčbu bakteriálních infekci nárůst rezistence bakterií ke konvenčním antibiotikům. Alternativní možností boje proti odolným bakteriím jsou enzymy s baktericidními účinky. Výhodou těchto enzymů je cílenost a vysoká efektivita proti rezistentním kmenům. Možnému klinickému využití zatím bráni jejich nízká stabilita a z ní plynoucí omezená doba antimikrobiální aktivity. Jedním z těchto „nadějných enzybiotik“ je lysostaphin, který cíleně rozkládá buněčnou stěnu patogenní bakterie Staphylococcus aureus. Ve své bakalářské práci se zabývám historií lysostaphinu, charakterizací baterie S. aureus, vlastnostmi vybraných lytických enzymů a především zvýšením/prodloužením stability lysostaphinu za pomocí proteinového inženýrství. V praktické části byla provedena příprava mutantního lysostaphinu, ověřena aktivita enzymu metodou lýze buněk na krevním agaru, testována stabilita proteinů pomocí metody založené na termodynamické denaturaci a následně porovnána stabilita původního a upraveného enzymu v závislosti na složení pufračního média a pH.

Abstract:

In the global view a treatment of bacterial infections is complicated by an increasing resistance of bacteria to conventional antibiotics. Alternative option of fight against resistant bacteria are enzymes with a bactericidal effect. An advantage is that these enzymes are targeted and highly effective against resistant strains. A clinical use is not possible yet for the low (insufficient) stability of these enzymes and therefrom resulting limited time of the antimicrobial activity. One of these promising enzybiotics is lysostaphin, which targetedly decomposes a cell wall of pathogenic bacteria Staphylococcus aureus. In my bachelor thesis, I deal with the history of lysostaphin, properties of selected lytic enzymes and mainly increasing or prolongation of the lysostaphin stability using the protein engineering. In the practical part, the mutant of lysostaphin was prepared, the activity of the enzyme was verified by the lysis of cells on a blood agar, the stability of proteins was tested using methods of thermodynamic denaturation and finally the stabilities of original and modified enzyme depending on the composition of the buffering medium and pH were compared.
