Small noncoding RNAs as diagnostic biomarkers for meningioma patients

Abstract:

Meningeomy tvoří více než jednu třetinu všech nádorů centrální nervové soustavy (CNS) a jedná se tak o nejpočetnější nádory CNS. Na základě histopatologické klasifikace definované WHO rozdělujeme meningeomy do tří skupin reflektujících současně jejich stupeň malignity a biologické chování. Zatímco nádory spadající do skupiny WHO I se vyznačují téměř výhradně benigním charakterem, se zvyšujícím se stupněm malignity narůstá také jejich agresivita. I přes významné pokroky v diagnostice meningeomů je stále obtížné predikovat zvýšené riziko rekurence nádoru po chirurgickém zákroku u pacientů s atypickým meningeomem (WHO grade II). MikroRNA (miRNA) jsou posttranskripční regulátory genové exprese. Tyto krátké nekódující RNA jsou zapojeny v kritických biologických procesech, včetně buněčné proliferace, diferenciace, apoptózy a imunitní odpovědi, a změna jejich exprese přispívá k patogenezi mnoha lidských malignit, včetně meningiomu. Studie také ukázaly jejich potenciál predikovat riziko rekurence atypických meningeomů. Cílem této práce bylo stanovení optimální referenční hladiny genu pro normalizaci RT-qPCR dat a následná identifikace expresního profilu miRNA v tkáni atypických meningeomů za použití technologie TLDA. Výsledkem této práce byla identifikace miR-142-5p a miR-338-5p jako potenciálních prognostických biomarkerů schopných predikovat rekurenci u pacientů s atypickým meningeomem.

Abstract:

Meningiomas make up more than one-third of all central nervous system (CNS) tumors and are the most common CNS tumors. Based on the histopathological classification defined by the WHO, we divide meningiomas into three groups that simultaneously reflect their degree of malignancy and biological behavior. While tumors belonging to the WHO I group are characterized almost exclusively by a benign character, their aggressiveness rises with increasing degree of malignancy. Despite significant advances in the diagnostics of meningiomas, it is still difficult to predict an increased risk of tumor recurrence after surgery in patients with atypical meningioma (WHO grade II). MicroRNAs (miRNAs) are post-transcriptional regulators of gene expression. These short non-coding RNAs are involved in critical biological processes, including cell proliferation, differentiation, apoptosis, immune responses, and altering their expression contributes to the pathogenesis of many human malignancies, including meningioma. Studies has also shown their potential to predict the risk of recurrence of atypical meningiomas. This work aimed to determine optimal reference gene for RT-qPCR normalization, identify miRNAs that occur in the atypical meningioma tissue using TLDA technology. This work resulted in identification of miR-142-5p and miR-338-5p as potential prognostic biomarkers with ability to predict recurrence of atypical meningioma.
