Monitoring of xylem and floem formation of European beech (Fagus sylvatica L.) at the cellular level in research site Rájec-Holíkov.

Abstract:

Předmětem závěrečné práce byl monitoring činnosti kambia vzorníkových stromů v letech 2013 a 2014. V závěrečné práci jsou shrnuty poznatky o průběhu kambiální aktivity, radiálním přírůstu kmene, procesu diferenciace buněk dřeva a tvorbě lýka. Pozorování vývoje kambiální aktivity na buněčné úrovni bylo umožněno díky zvolené metodě periodických odběrů. Periodické odběry byly realizovány pomocí nástroje Trephor v týdenních intervalech od března do listopadu. Z odebraných vzorků byly zhotoveny trvalé mikroskopické preparáty zachycující informaci o aktuální činnosti kambia v okamžiku odběru. Průběh a výsledky kambiální aktivity byly posuzovány ve vztahu ke klimatickým podmínkám lokality. Nepříznivý vývoj klimatických podmínek byl zaznamenán v růstové reakci vzorníkových stromů zejména v roce 2014, kdy byl vytvořen letokruh o celkové šířce 1012±489 µm. Maximální denní přírůst xylému 13±4 µm byl zaznamenán v období od 29. 5. do 26. 6. 2014. V roce 2013 dosahovala celková šířka letokruhu 1655±444 µm s maximálním denním přírůstem 27±13 µm v období od 13. 6. 2013 do 11. 7. 2013. Tvorba nových buněk xylému trvala v roce 2013 průměrně 91 dní. V následujícím roce 2014 bylo období produkce nových buněk xylému o 9 dní kratší.

Abstract:

Monitoring of cambial activity in sample trees during growing season 2013 and 2014 was the aim of this thesis. This study summarizes the findings about the variations in the cambial zone, the radial increment of stems, the differentiation process of xylem and phloem formation. The changes in cambial zone observed at the cellular level were made possible by the periodical sampling. Periodical samples - microcores were taken with the Trephor tool at weekly intervals from March to November. The permanent microscope slides were made of microcores. The number of the permanent microscope slides provided information about the actual cambial activity at the time of sampling. Variations and results of the cambial activity were evaluated in relation to the climatic conditions of the research site. The unfavorable climatic conditions were noted in the growth response of sample trees, especially in 2014. The radial xylem increment was 1012±489 µm in 2014. The maximum daily increment of xylem 13±4 µm was recorded between 29 May and 26 June 2014. The total tree ring width was 1655±444 µm in 2013. The maximum daily increment of xylem 27±13 µm was recorded between 13 June and 11 July 2013. The xylem formation lasted for 91 days in 2013. The xylem formation was 9 days shorter in 2014.
