Abstract:

Táto bakalárska práca sa zaoberá vývojom nového syntetického postupu pre modifikáciou betulínu v polohe C-30. Pre prípravu knižnice derivátov bol použitý nasledovný syntetický plán. Prvým krokom bolo zavedenie vhodných chrániacich skupín do skeletu betulínu v polohách C-3 a C-28, následne bola poloha C-30 oxidovaná na aldehyd. Jeho reakciou s organokovovou zlúčeninou obsahujúcou trojitú väzbu a následnou oxidáciou bol získaný triterpenoidný alkín vhodný pre Huisgenove 1,3-dipolárne cykloadície s organickými azidmi. V rámci tejto bakalárskej práce boli pripravené dva cieľové deriváty. Hlavný prínos tejto práce spočíva v preverení a optimalizácii celej syntetickej cesty, vrátane charakterizácii medziproduktov a cieľových zlúčenín pomocou bežných spektrálnych dát (NMR, IČ, HRMS). Táto bakalárska práca sa člení na niekoľko častí. Teoretická časť obsahuje charakteristiku terpénov, terpenoidov, ich delenie a syntézu. Taktiež popisuje betulín a bioizostériu. Diskusná časť rozoberá problematiku, ktorá vznikla pri jednotlivých reakciách. Experimentálna časť popisuje presné reakčné postupy, analýzu a charakterizáciu produktov.

Abstract:

This bachelor thesis deals with the development of a new synthetic procedure for the modification of betulin at the C-30 position. The following synthetic scheme was used for the preparation of the derivative library. The first step was the introduction of suitable protecting groups into the betulin skeleton at the C-3 and C-28 positions, followed by oxidation of the C-30 position to aldehyde. Its reaction with an organometallic compound containing a triple bond and subsequent oxidation afforded a triterpenoid alkyne suitable for Huisgen 1,3-dipolar cycloadditions with organic azides. Two target derivatives were prepared in the framework of this bachelor thesis. The main contribution of this work lies in the validation and optimization of the whole synthetic route, including the characterization of intermediates and target compounds using conventional spectral data (NMR, IR, HRMS). This bachelor thesis is divided into several parts. The theoretical part includes the characterization of terpenes, terpenoids, their partitioning and synthesis. It also describes betulin and bioisostery. The discussion part discusses the issues that arose in the various reactions. The experimental part describes the exact reaction procedures, analysis and characterization of the products.
