Ecotoxicity of complex cyanobacterial biomass and its components

Abstract:

Potřeba studovat mechanismy toxicity sinicového vodního květu je zapříčiněna jeho masovým rozvojem a negativním vlivem na živé organismy. Účinky známých toxinů cyanobakterií (zvláště microcystinů) byly intenzivně zkoumány, řada studií však naznačuje, že toxicita biomasy sinic může být zapříčiněna neznámými cyanotoxiny, jejich spolupůsobením mezi sebou, se známými toxiny, případně s částmi buněk. Významný vliv na celkovou toxicitu biomasy sinic mohou mít ale také takové parametry, jako koncentrace rozpuštěného kyslíku, amoniaku apod. V praktické části byla porovnávána toxicita pěti frakcí cyanobakteriální biomasy (komplexní biomasa, pelet, supernatant, permeát a eluát). K tomu byl použit akutní test toxicity na Daphnia magna. Nejvýraznější negativní vliv byl sledován u komplexní biomasy a pak peletu a supernatantu. Ale eluát (obsahující hlavně microcystiny) se ukázal jako netoxický. Tyto výsledky mohou být způsobeny neznámými toxiny a spolupůsobením látek. Dle měření však zásadní roli v celkovém negativním účinku sinicové biomasy měl pokles koncentrace rozpuštěného kyslíku. Paralelně byl prováděn test na buněčné linii H4IIEluc (frakce supernatant, permeát, eluát), kde se supernatant ukázal jako toxičtější než ostatní dvě frakce, což indikuje možné spolupůsobení látek v této frakci.

Abstract:

The urgent need for study of the toxic mechanisms of cyanobacterial water bloom results from the mass development of cyanobacteria in aquatic environments and its negative effects on living organisms. The effects of known cyanotoxins (especially microcystin) have been investigated intensively, however, a lot of studies indicate that the overall toxicity of cyanobacterial biomass may be caused by unknown cyanotoxins, their interactions as well as their interactions with the known cyanotoxins or various cell parts. Moreover, the overall toxicity is significantly affected by conditions such as the level of dissolved oxygen and ammonia. In the practical part of the bachelor thesis, toxicity of five fractions of cyanobacterial biomass (complex biomass, pellet, aqueous extract, permeate and eluate) was compared. The acute test on Daphnia magna was used as means of assessing the toxicity of these five fractions. The most significant adverse effect was observed in the case of complex biomass followed by pellet and aqueous extract while eluate (containing mainly microcystin) was nontoxic. These results may suggest the presence of unknown toxins and joint interactions of contained substances. However, the measurements showed that low levels of dissolved oxygen played dominant role in the overall negative effect of cyanobacterial biomass. At the same time, the aqueous extract, permeate and eluate were also tested on the cell line H4IIEluc where the aqueous extract showed the greatest toxicity indicating interactions of contained substances.
