Molecular-phylogenetic and taxonomic studies of selected taxa of Bibionomorpha (Diptera)

Abstract:

The infraorder Bibionomorpha belongs to the most numerous groups of two-winged flies (Diptera), mainly due to the superfamily Sciaroidea. Within this superfamily, families Cecidomyiidae, Mycetophilidae and Sciaridae clearly dominate by the number of species. The presented dissertation summarizes the current knowledge about the classification, relationships and biology of the infraorder Bibionomorpha and the superfamily Sciaroidea, focusing on the families Mycetophilidae, Keroplatidae and Sciaridae. The aim of this dissertation is to reconstruct the molecular phylogeny of selected taxa of the superfamily Sciaroidea. The analyses were conducted based on various combinations of mitochondrial (12S, 16S, COI, COII, cytB) and nuclear (18S, 28S, ITS2, CAD, MCS, EF1?) gene markers. Reconstructions of molecular phylogeny of the families Keroplatidae and Ditomyiidae, as well as of the subfamilies Sciophilinae and Leiinae (family Mycetophilidae) and the Sciaroidea incertae sedis, were performed for the first time. Molecular phylogeny of the family Sciaridae, that of the tribe Exechiini (Mycetophilidae, subfamily Mycetophilinae), genera Docosia Winnertz, 1863 (Mycetophilidae, subfamily Gnoristinae) and Catocha Haliday, 1833 (family Cecidomyiidae) were also studied. Results are presented in five published papers and in one unpublished manuscript included in this dissertation. Twenty-nine new species were described within these published papers.

Abstract:

Infrařád Bibionomorpha patří mezi druhově nejpočetnější skupiny dvoukřídlého hmyzu, a to zejména díky nadčeledi Sciaroidea. V rámci této nadčeledi jednoznačně dominují počtem druhů čeledi Cecidomyiidae, Mycetophilidae a Sciaridae. V předložené dizertační práci jsou shrnuty dosavadní poznatky o klasifikaci, příbuzenských vztazích a biologii infrařádu Bibionomorpha a nadčeledi Sciaroidea, se zaměřením na čeledi Mycetophilidae, Keroplatidae a Sciaridae. Cílem této disertační práce je rekonstruovat molekulární fylogenezi vybraných taxonů nadčeledi Sciaroidea. Molekulární fylogeneze byly provedeny na základě různých kombinací mitochondriálních (12S, 16S, COI, COII, cytB) a jaderných (18S, 28S, ITS2, CAD, MCS, EF1?) genových markerů. Vůbec poprvé byly provedeny rekonstrukce molekulární fylogeneze čeledí Keroplatidae a Ditomyiidae a také podčeledí Sciophilinae a Leiinae (čeleď Mycetophilidae) a skupiny Sciaroidea incertae sedis. Dále byly uskutečněny studie věnované molekulární fylogenezi čeledi Sciaridae, tribu Exechiini (Mycetophilidae, podčeleď Mycetophilinae), rodů Docosia Winnertz, 1863 (Mycetophilidae, podčeleď Gnoristinae) a Catocha Haliday, 1833 (čeleď Cecidomyiidae). Výsledky jsou uvedeny v pěti publikovaných článcích a jednom nepublikovaném rukopise, které jsou součástí této disertační práce. V rámci těchto publikovaných článků bylo popsáno 29 nových druhů.
