Study of Carbon Monoxide-Releasing Flavonols Using Spectroscopic Methods

Abstract:

Tato práce se zabývá návrhem, syntézou a studiem mechanismu fotochemických a fotofyzikálních procesů látek, které jsou schopny uvolňovat oxid uhelnatý po ozáření světlem. Shrnuté výsledky již byly publikovány nebo budou zaslány k publikaci v blízké budoucnosti. Dizertační práce je rozdělena do tří kapitol: literární rešerše, výsledky a diskuze a experimentální část. Literární rešerše shrnuje již publikované informace relevantní k zkoumaným projektům, jako je popis fotofyzikálních a fotochemických procesů, tranzientní spektroskopie, biologických efektů oxidu uhelnatého a fotofyziky a fotochemie flavonoidů a cyaninových barviv. Kapitola Výsledky a diskuze je dále rozdělena do čtyř částí, ve kterých jsou diskutovány konkrétní projekty. První část se zabývá mechanistickou studií uvolňování oxidu uhelnatého z látek založených na 3-hydroxyflavonu. Diskutuji zde multiplicitu produktivního excitovaného stavu a roli kyslíku při uvolňování oxidu uhelnatého, odhalujíc tak dichotomii fotoreakce kyselé a bazické formy 3-hydroxyflavonu. Druhá část popisuje systematickou studii vztahu struktury a reaktivity heptamethinových cyaninových barviv nesoucích substituenty v různých polohách. Ukazuji, že jak druh, tak i pozice substituentu významně ovlivňují vlastnosti cyaninových barviv. Třetí část představuje koncept spojení struktury flavonolu s NIR-absorbujícím chromoforem, konkrétně cyaninem, které tvoří novou třídu látek uvolňující oxid uhelnatý po ozáření světlem. Čtvrtá část popisuje obsáhlý výzkum substituovaných flavonolů s cílem vylepšit jejich fotoreaktivitu, aplikovatelnost v biologii a uvolňování oxidu uhelnatého. Použité materiály a metody jsou detailně popsány v třetí kapitole. Publikované články a experimentální data jsou doplněny v příloze.

Abstract:

This Ph.D. work is focused on the design, synthesis, and mechanistic investigation of photochemical and photophysical processes on some carbon monoxide-releasing molecules (photoCORMs). The results summarized in this thesis have either been published or will be submitted for publication in the near future. The thesis is divided into three main parts: a literature part, results and discussion, and an experimental part. The literature part summarizes the previously published information relevant for the studied projects, such as the description of photophysical and photochemical processes, transient spectroscopy, biological effects of carbon monoxide, and photophysics and photochemistry of flavonoids and cyanine dyes. The chapter Results and Discussion is divided into four paragraphs, in which four different projects are discussed. The first part deals with mechanistic studies of CO release from a 3-hydroxyflavone based motif. I address the multiplicity of the productive excited states and the role of oxygen in the CO liberation, revealing a photoreaction dichotomy of the 3-hydroxyflavone acid and base forms. The second part is about the systematic studies of the structure and property relations in heptamethine cyanines, bearing different substituents in different positions. It is demonstrated that both the type and the position of a substituent significantly influence the properties of these cyanine dyes. The third part introduces the concept of fusion of the flavonol motif with a NIR-absorbing chromophore, specifically heptamethine cyanine, to obtain a new class of transition-metal-free photoCORMs. The fourth part describes the comprehensive investigation of differently substituted flavonols, aiming to improve the CO release and their photoreactivity and bioapplicability. The publications and experimental data are appended to the last part of the thesis.
