Redox processes of electroactive amino acids

Abstract:

Bakalářská práce se zabývá studiem elektrochemické aktivity některých aminokyselin na grafitových elektrodách. Průzkum voltametrických oxidačních signálů L-tyrosinu, L-tryptofanu, L-cysteinu, L-methioninu a L-histidinu na nemodifikovaném grafitovém substrátu čtyřech různých elektrod ukázal, že nejvhodnější a nejcitlivější elektroanalýzu lze očekávat na polymerní pentelkové elektrodě (pPeGE). Ze všech aminokyselin jenom L-tyrosin, L-tryptofan a L-cystein poskytovaly čitelné oxidační voltametrické signály. Zatímco signály L-tyrosinu a L-tryptofanu se často překrývají, oxidační pík L cysteinu je na potenciálové škále dostatečně od těchto dvou aminokyselin odlehlý. Proto byly podrobně sledovány především oxidační procesy L-tyrosinu a L-tryptofanu. Bylo zjištěno, že při určitých hodnotách pH by měla být separace tyrosinových a tryptofanových oxidačních píků sice reálná, ale silně ovlivněná jejich koncentračním poměrem. Bakalářská práce přináší nové užitečné poznatky týkající se nejen elektroanalýzy tyrosinu a tryptofanu na grafitových elektrodách a jejich mechanistických procesů, ale i další informace použitelné při řešení problémů nativních a denaturačních změn proteinů, které elektroaktivní aminokyseliny obsahují.

Abstract:

This bachelor thesis deals with the study of the electrochemical activity of several amino acids on graphite electrodes. The survey of voltammetric oxidation signals of L tyrosine, L-tryptophan, L-cysteine, L-methionine, and L-histidine on four types of unmodified graphie electrode surfaces has shown that the most suitable and sensitive electroanalysis can be expected on the polymer pencil graphite electrode (pPeGE). Out of all these amino acids, only L-tyrosine, L-tryptophan, and L-cysteine provided clear oxidation signals. However, the peaks of L-tyrosine and L-tryptophan often overlap, while the peak of L-cysteine is sufficiently separated. Therefore, the oxidation processes of these two amino acids were studied in greater detail. It was found that at certain pH values, the separation of the L-tyrosine and L-tryptophan peaks was real, but strongly affected by their concentration ratio. This thesis brings new useful information related to not only the electroanalysis of L-tyrosine and L-tryptophan on graphite electrodes, but also additional information applicable in the solution of problems of native and denaturation changes of proteins, which contain these electroactive amino acids.
