The comparison gene aktivity of MMPs during wound healing by healthy and diabetic rats

Abstract:

Matrixové metaloproteinázy (MMP) jsou enzymy, které proteolyticky degradují extracelulární matrix (ECM) a tím jsou odpovědné za remodelaci pojivové tkáně. Za fyziologických podmínek se podílejí na ontogenetickém vývoji, morfogenezi, angiogenezi, růstu a hojení ran. Objevují se také ve většině zánětlivých, degenerativních a maligních procesech. Hojení je komplikovaný děj, kterého se kromě MMP účastní celá řada buněk (neutrofily, lymfocyty, mastocyty, monocyty, makrofágy, fibroblasty, keratinocyty, endotel) a jimi vylučovaných působků (cytokiny, růstové faktory apod.). Tento děj probíhá v několika fázích: hemostáza, zánět, proliferační fáze, fibrotizace a tvorba jizvy. V této diplomové práci jsme se zaměřili na sledování změn aktivit vybraných MMP a jejich inhibitorů (TIMP) v průběhu hojení ran u zdravých a diabetických potkanů. Modelovými zvířaty byli zvoleni potkani Zucker Diabetic Fatty (ZDF), kterým byly odebírány v časových intervalech vzorky kůže, granulační tkáně a jizvy. Z těchto vzorků byla dále izolována celková RNA a syntetizována cDNA. Exprese genů byla stanovena metodu real-time PCR. Data byla statisticky zpracována pomocí Studentova t-testu (p ? 0,05). MMP zasahují do všech fází hojení a jejich exprese se v průběhu hojení mění. V neporušené kůži byly lehce zvýšené hodnoty exprese MMP-2, -3, -14 a TIMP-2. První den exprese prudce vzrostla u MMP-8, -9 a TIMP-1 a vzápětí prudce klesla. Od pátého dne hojení narůstá exprese MMP-13, -2, -14 a TIMP-2. Desátý den je exprese vysoká u MMP-13, -2, -10, -14, -12 a TIMP-2. I v čerstvé zahojené jizvě lze zaznamenat vyšší expresi MMP-2, -10, -13 -14 a TIMP-2. Nezaznamenali jsme žádné významné rozdíly v expresi sledovaných genů mezi zdravými a diabetickými zvířaty až na výjimku v neporušené kůži, kde byla hodnota exprese MMP-3 u samců. Při porovnání samců a samic byly hodnoty exprese MMP-2, MMP-14 a TIMP-2 u samců vyšší v neporušené kůži, naopak 3. den hojení byly u samic zvýšené hodnoty exprese MMP-14 a TIMP-2.

Abstract:

Matrix metalloproteinases (MMPs) are enzymes that proteolytically degrade extracellular matrix (ECM) and they are thus responsible for the remodelling of connective tissue. MMPs involve in the ontogenesis, morphogenesis, angiogenesis, cell growth and wound healing in the physiological conditions. They take part in inflammation, degenerative and malignant processes. Healing is a very complicated process. MMPs and many chemokines (cytokines, growth factors, proteases) from cells (neutrophils, lymphocytes, mast cells, monocytes, macrophages, fibroblasts, keratinocytes, and endothelium) involve in the wound healing. This process takes place in several phases: hemostasis, inflammation, proliferative phase, and the formation of fibrotic scars. We focused on monitoring expressions of selected MMPs and their inhibitors (TIMP) during wound healing in healthy and diabetic rats in this thesis. Zucker Diabetic Fatty rat (ZDF) has been chosen as a suitable model for our study. We used the real-time RT-PCR methods for study expressions of MMPs and their inhibitors. Data were statistically analyzed using Student's t-test (p ? 0.05). MMPs are produced by different cell types in the healing and by expression of genes changed over time. The value of expression MMP-2, -3, -14 and TIMP-2 was slightly elevated in the intact skin. The expression of MMP-8, -9 and TIMP-1 rapidly increased the first day and then dramatically decreased. From the fifth day of healing the expression of MMP-13, -2, -14 and TIMP-2 increases. The high expressions of MMP-13, -2, -10, -14, -12 and TIMP-2 appeared on in the tenth day. We can see a higher expression of MMP-2, -10, -13 -14 and TIMP-2 even in fresh healed scar. We did not reveal any significant differences in gene expression between healthy and diabetic animals with an exception in the intact skin, where the value of MMP-3 increased by diabetics. The values of expression MMP-2, MMP-14 and TIMP-2 in intact skin were increased at males, but the increased value of expression MMP-14 and TIMP-2 were observed in female on the third day of the healing.
