Effect of chlorine and other biotechnologically manufactured antiseptics on the activation of phagocytosis

Abstract:

Fagocytóza je buněčný proces, který se uplatňuje při obranných reakcích imunitního systému, ale také při hojení ran a regenerace tkáně. Hojení akutní rány zahrnuje několik fází, ve kterých se nejprve v poškozené tkáni objevují neutrofily a následně se zvyšuje také počet zánětlivých monocytů a makrofágů. Antiseptické látky jsou užívány ke snížení bakteriální zátěže například chronických infikovaných ran. Vedlejší efekty antiseptik a jejich vliv na procesy probíhající v ranách se zkoumají. Cílem diplomové práce bylo stanovení vlivu vybraných antiseptických látek na aktivaci fagocytózy u lidské buněčné linie THP-1 makrofágů a lidských neutrofilů (PMN). Byla použita antiseptika vykazující oxidační aktivitu a působící skrz halogeny: N-chlorotaurin (NCT), jod (PVP-I), chlornan sodný a chloramid hyaluronanu (HACl); a antiseptika působící jinými mechanismy: chlorhexidin dihydrochlorid (CHX), oktenidin dihydrochlorid (OCT), polyhexamethylen biguanid hydrochlorid (PHMB) a dusičnan stříbrný. Antiseptická látka HACl byla připravena biotechnologickou cestou. Vliv antiseptik na zvýšení fagocytózy nebyl pozorován u žádné z testovaných látek.

Abstract:

Phagocytosis is a cellular process which takes place during defence reactions of immune system, but also during wound healing and tissue regeneration. Acute wound healing includes several phases. First neutrophils appear in the damage tissue, and then the amount of inflammatory monocytes and macrophages also increases. Antiseptics are used to reduce the bacterial load of, for example, chronic infected wounds. The side effects of antiseptics and their effect on ongoing wound processes are examined. The aim of this diploma thesis was to determine the effect of selected antiseptics on the activation of phagocytosis of human cell line THP-1 macrophages and human neutrophils (PMNs). Antiseptics with an oxidation effect and antiseptics acting through halogens were used: N-chlorotaurine (NCT), iodine (PVP-I), sodium hypochlorite and hyaluronic acid chloramide (HACl). Antiseptics using different mechanisms were also used: chlorhexidine dihydrochloride (CHX), octenidine dihydrochloride (OCT), polyhexamethylene biguanide hydrochloride (PHMB) and silver nitrate. Antiseptic HACl was produced by a biotechnological pathway. No effect on increasing phagocytosis was observed for any of the tested antiseptics.
