Male flower induction in paternal lines of parthenocarpic cucumber

Abstract:

Tato diplomová práce se věnovala problematice hledání environmentálně bezpečných látek alternativních ke standardně používanému dusičnanu stříbrnému ve vztahu k možnosti navození produkce samčích květů u odrůd partenokarpických okurek. Celosvětový trend hledání udržitelných způsobů života lidí na planetě Zemi se promítá i do zemědělského výzkumu. Laboratorní látky jsou často reagenciemi toxickými pro živé organismy včetně člověka, či environmentálně nebezpečné látky, které se postupně kumulují v organismu a způsobují postupné chřadnutí či reprodukční problémy. Dusičnan stříbrný (AgNO3) je látkou vysoce toxickou a environmentálně nebezpečnou. Zároveň se jedná o vysoce účinný stimulant tvorby samčích květů u partenokarpických okurek, standardně využívaný ve šlechtitelství okurek. V praktické experimentální části bylo testování podrobeno využití stříbrných nanočástic (AgNPs), od kterých si výzkumníci slibují nižší environmentální riziko, dále látek běžně využívaných jako hnojiva (Na3PO4, KNO3) a vystavení testovaných rostlin podchlazení za účelem stimulace tvorby samčích květů na partenokarpických odrůdách okurek. Firma MORAVOSEED CZ a. s., se sídlem v Mikulově na Moravě, poskytla k experimentu dvě své odrůdy AL a SAN. Sadba těchto odrůd byla připravena pracovníky firmy MORAVOSEED CZ a. s. a poté byla předána k experimentálním ošetřením rostlin. Ošetření rostlin dusičnanem stříbrným (AgNO3) představovalo dle předpokladu velmi účinnou variantu ovlivnění míry tvorby samčích květů na okurkách. Z ostatních ošetření, u nichž byl zkoumán potenciál vyvolání stejného efektu se nejperspektivnějším ukázalo ošetření rostlin dusičnanem draselným (KNO3). Molekulárními analýzy pak poukázaly na účinnost ošetření rostlin hnojivy (KNO3, Na3PO4) v kontextu snížení míry genové exprese v ethylénové dráze rostlin, což je jeden z předpokladů indukce tvorby samčích květů. Ten se však při měření počtu samčích květů na rostlinách potvrdil pouze u variant ošetřených KNO3. Perspektivita využití hnojiv k indukci tvorby samčích květů a jejich využití jakožto environmentálně bezpečnější alternativ k běžně užívanému dusičnanu stříbrnému se v případě KNO3 v rámci výzkumu provedeného v této diplomové práci prokázala. Nyní je navrhována optimalizace metodiky využití dusičnanu draselného (KNO3) k indukci tvorby samčích květů u partenokarpických odrůd okurek.

Abstract:

In this thesis, the problem of searching for environmentally safe alternatives to the standard silver nitrate in relation to the possibility of inducing male flower production in parthenocarpic cucumber varieties was addressed. The worldwide trend of seeking sustainable ways of living on planet Earth is also reflected in agricultural research. Laboratory substances are often reagents that are toxic to living organisms, including humans, or environmentally hazardous substances that accumulate in the body over time and cause gradual wasting or reproductive problems. Silver nitrate (AgNO3) is a highly toxic and environmentally hazardous substance. It is also a highly effective stimulant of male flower formation in parthenocarpic cucumbers, and is standard in cucumber breeding. In the practical experimental part, the use of silver nanoparticles (AgNPs), from which the researchers expect lower environmental risk, as well as substances commonly used as fertilizers (Na3PO4, KNO3) and exposure of the tested plants to hypothermia to stimulate the formation of male flowers on parthenocarpic cucumber varieties were tested. MORAVOSEED CZ a. s., based in Mikulov na Moravě, provided two of its varieties AL and SAN for the experiment. The seedlings of these varieties were prepared by MORAVOSEED CZ a. s. workers and then handed over for experimental plant treatments. The treatment of plants with silver nitrate (AgNO3) was expected to be a very effective option for influencing the rate of male flower formation on cucumber plants. Of the other treatments that were investigated for their potential to induce the same effect, potassium nitrate (KNO3) treatment proved to be the most promising. Molecular analyses then indicated the efficacy of plant fertilizer treatments (KNO3, Na3PO4) in the context of a reduction in the level of gene expression in the ethylene pathway of plants, which is one of the prerequisites for the induction of male flower formation. However, this was only confirmed in the KNO3-treated variants when measuring the number of male flowers on the plants. The prospect of using fertilizers to induce male flower formation and their use as environmentally safer alternatives to the commonly used silver nitrate was demonstrated in the case of KNO3 in the research carried out in this thesis. Optimisation of the methodology of using potassium nitrate (KNO3) to induce male flower formation in parthenocarpic cucumber varieties is now proposed.
