Non-canonical role of telomerase complex in hematopoietic differentiation and the immune system

Abstract:

Telomeropatie patří mezi vzácná onemocnění asociovaná s poruchou molekulárních mechanismů kontrolujících délku telomér. Telomeropatie jsou nejčastěji důsledkem genetických mutací telomerázového komplexu. Dyskeratosis kongenita je vrozená progresivní telomeropatie s mutací v sekvenci pro “telomerase RNA component” (TERC), která vede k narušení schopnosti udržování délky telomér a k následnému buněčnému stárnutí a klinickým projevům zrychleného stárnutí. Nejzávažnějším klinickým projevem nemoci je narušení krvetvorby a selhání kostní dřeně vedoucí k imunodeficitům, nicméně mechanismus, který tento proces způsobuje není zatím dostatečně pochopen. V této práci jsme vyvinuli model poruchy telomerázové funkce tím, že jsme pomocí shRNA vytvořili knockdown TERC exprese v lidských indukovaných pluripotentních buňkách (iPSCs). Potom jsme v iPSCs buňkách stimulovali in vitro krvetvorbu, během které jsme analyzovali vliv potlačeného TERC. Snížená exprese TERC vyvolala změny v diferenciaci hematopoetických buněk a zvýšila expresi znaků buněčné senescence a přítomnost reaktivních forem kyslíku (ROS). Je zajímavé, že délka telomer zůstala u shTERC knockoutovaných iPSCs nezměněná, současně buňky zvýšily mRNA expresi a aktivitu telomerázové podjednotky (TERT). Dále jsme se zaměřili na efekt TERC deplece v THP-1 buněčné linii, kde jsme také pozorovali sníženou schopnost diferenciace. Oba typy myeloidních buněk diferenciovaných z shTERC-iPSCs i z THP1 měly sníženou kapacitu fagocytózy a zvýšenou úroveň apoptózy. Tato data byla potvrzena použitím inhibitoru TERT aktivity (BIBR1532) na kultuře primárních HSCs, to naznačuje, že tento fenotyp je kontrolován netelomerickou rolí telomerázy. Tato data umožnují lepší pochopení procesů, které řídí důležité klinické příznaky telomeropatií, a dále identifikují nové funkce telomerázového komplexu mimo vlastní kontrolu délky telomér. Dále jsme chtěli pochopit, zdali stejný molekulární mechanismus může způsobovat i poškození jiných tkání, ke kterému během telomeropatií dochází, a také zdali telomerázová aktivita může být ovlivněna zánětem. Za tímto účelem jsme použili model střevních organoidů diferenciovaných z iPSCs buněk. Zánětlivá odpověď buněk střevních organoidů byla vyvolána stimulací PRR ligandy, především LPS, Poly I:C a tepelně inaktivovanými konidiemi. Použitím TERT inhibitoru (BIBR1532) jsme ukázali, že exprese zánětlivých cytokinů IL-1β, IL-8, a TNF-α a prozánětlivého znaku MMP9 jsou sníženy v závislosti na blokované aktivitě telomerázy. Tento výsledek potvrzuje netelomerickou funkci telomerázy. Tato práce používá nové buněčné modely vedoucí ke zjištění, že telomerázová aktivita má přímý vliv na krvetvorbu a na funkce imunitního systému včetně fagocytózy a produkce cytokinů. Nyní jsou potřeba další studie, které vysvětlí detaily nekanonické role telomerázy, které jsou asociovány s telomeropatiemi, a případně identifikují možnosti terapeutické intervence.

Abstract:

Telomeropathies are rare disorders associated with impaired telomere length control mechanisms that frequently result from genetic mutations in the telomerase complex. Dyskeratosis congenita is a congenital progressive telomeropathy in which mutation in the telomerase RNA component (TERC) impairs telomere maintenance leading to accelerated cellular senescence and clinical outcomes resembling premature aging. The most severe clinical feature is perturbed hematopoiesis and bone-marrow failure, leading to immunodeficiency, but the underlying mechanisms are not fully understood. Here, we developed a model of telomerase function imbalance using shRNA to knockdown TERC expression in human induced pluripotent stem cells (iPSCs). We then promoted in vitro hematopoiesis in these cells to analyse the effects of TERC impairment. Reduced TERC expression impaired hematopoietic stem-cell (HSC) differentiation and increased the expression of cellular senescence markers and production of reactive oxygen species. Interestingly, telomere length was unaffected in shTERC-iPSCs. shTERC-iPSCs also had upregulated TERT mRNA level and activity. We then assessed the effects of TERC-depletion in THP-1 myeloid cells and again observed reduced hematopoietic and myelopoietic differentiating potential. Both shTERC-knockdown iPSC-derived and THP-1-derived myeloid cells had lower phagocytic capacity and higher apoptosis. These findings were confirmed using a BIBR1532 TERT inhibitor on primary HSCs, suggesting that the phenotype is controlled by non-telomere functions of telomerase. These data provide a better understanding of the molecular processes driving the clinical signs of telomeropathies and identify novel roles of the telomerase complex other than regulating telomere length. In order to further understand whether similar molecular mechanism affect different tissues susceptible to telomeropathies and the crosstalk between telomerase and inflammation, we use the model of iPSC-derived gut organoid. Inflammation was induced in the intestinal organoid by stimulation using PRR ligands, including LPS, Poly I:C, and heat inactivated conidia. Using the TERT inhibitor, BIBR1532, we show that the expression of inflammatory cytokines IL-1β, IL-8, and TNF-α and pro-inflammatory marker MMP9 are reduced upon telomerase inhibition in intestinal organoids, indicating a non-telomere function of telomerase. The study uses innovative cellular models to conclude that telomerase activity has a direct role in hematopoiesis and immune system functions, including phagocytosis and cytokine production. Future studies are now needed to further dissect the non-canonical role of telomerase associated with telomeropathies and identify new therapeutic targets.
