Determination of carboxypeptidase B1 substrate specifity using C-TAILS approach

Abstract:

This bachelor thesis is focused on the substrate specificity of carboxypeptidase B1 (CPB1) using the C-Terminal Amine-based Isotope Labeling of Substrates (C-TAILS). CPB1, a tissue specific pancreatic enzyme, was recently identified as a potential biomarker of metastasis in luminal A breast carcinoma. In the experimental part of this thesis, we apply the C-TAILS method to determine the mechanisms of CPB1 activity. This method, utilizing mass spectrometry detection of peptides, represents an optimized methodology to identify and quantify C-terminal peptides under various experimental conditions. Comparison of C-terminal peptides from MCF-7 cell line with overexpressed CPB1 and control cell line allows identification of peptides cleaved by CPB1. Using these approaches, we quantified two peptides which originate from serotransferrin and cofilin. These peptides show statistically significantly increased levels in cell line with overexpressed CPB1 compared to control. The peptide sequences were located up to 10 amino acids from C-terminus of the proteins. In conclusion, it seems that serotransferrin and cofilin could be substrates of CPB1.

Abstract:

Bakalárska práca je zameraná na enzým karboxypeptidázu B1 (CPB1) a stanovenie jej substrátovej špecifity metódou izotopového značenia substrátov na C-terminálnom amíne (C-TAILS). CPB1 je tkanivovo špecifický enzým pankreasu, ale nedávne poznatky naznačujú, že by mohol hrať úlohu pri rozvoji metastáz do lymfatických uzlín u karcinómu prsníka subtypu luminal A. Pre objasnenie jej pôsobenia v tomto type buniek bude v experimentálnej časti využitá metóda C-TAILS. Táto metóda umožňuje v kombinácii s hmotnostnou spektrometriou identifikovať a kvantifikovať C-koncové peptidy proteínov a porovnaním s negatívnou kontrolou je možné určiť, ktoré proteíny CPB1 štiepi. Využitím týchto metód sa podarilo kvantifikovať dva peptidy patriace proteínom serotransferín a kofilín, ktoré vykazovali štatisticky významne zvýšené hladiny v línii s overexprimovanou CPB1 oproti kontrole a ich sekvencie sa v rámci proteínu nachádzali do 10 aminokyselín od C-konca. Na základe týchto výsledkov je možné usúdiť, že serotransferín a kofilín by mohli byť substrátmi pre CPB1.
