Methods of targeted genetic manipulations in algae

Abstract:

Cílem mé práce je shrnout a zpracovat problematiku metod pro cílenou genetickou manipulaci u řas. Cílené genetické manipulace jsou klíčem k porozumění funkcí genů u všech organismů. U řas, což jsou fototrofní organismy, se genetické manipulace využívají pro studium fotosyntézy. Dalšími oblastmi využití cílených genetických manipulací u řas jsou biotechnologie a medicína, kdy se vnášejí specifické geny do buňky, která následně syntetizuje jimi kódované proteiny dle našeho zájmu. Základem těchto manipulací je vyvolání dvojitých zlomů na DNA (tzv. DSB), které mohou být opraveny homologní rekombinací (HR) nebo nehomologním spojením konců (NHEJ), kdy je možno vyřadit nějaký gen z provozu nebo do DNA integrovat pro nás zajímavý gen. Právě k vhodné expresi těchto inzertovaných genů se využívají specifické promotory, řídící expresi onoho genu. Jelikož bývá úspěšnost těchto manipulací velice nízká, využívají se rozličné selekční markery a reportérové geny, umožňující selekci mutantů na určitém médiu nebo jejich rozpoznání pomocí látky, kterou produkují. V této práci nastíním aktuání klasifikaci řas a komplexně shrnu poznatky o metodách pro jejich cílenou genetickou manipulaci. Rozeberu využití promotorů, selekčních markerů a reportérových genů, a v neposlední řadě shrnu také využití metod pro genovou manipulaci, jako jsou ZFN, TALEN, CRISPR/Cas9 systém nebo RNA interference.

Abstract:

The aim of my thesis is to sum up and to compile the issue of methods for targeted genetic manipulation in algae. Targeted genetic manipulations are the key to understanding functions of genes for all the organisms. For algae, which are phototrophic organisms, are genetic manipulations used for studies of photosynthesis. Further, we can use targeted genetic manipulations of algae in biotechnology and medicine as we can bring specific genes into the cell, which later synthesizes any genes-encoded proteins we desire. Main principle of these manipulations is to cause double strand breaks of DNA, which can be fixed by homologous recombination or non-homologous end joining to inactivate target gene or to integrate some gene we find interesting to DNA. For feasible expression of inserted genes we use specific promoters controlling their expression. As we know that success of these manipulations is very low, we tend to use various selection markers and reporter genes, which facilitate selection of the mutants on specific medium or their recognition by the substance they produce. In this thesis I am going to outline current classification of algae and completely sum up knowledge of methods used for their targeted genetic manipulations. I am also going to analyze the use of promoters, selection markers and reporter genes and last but not least to summarize utilization of methods for genetic manipulation, such as ZFN, TALEN, CRISPR/Cas9 system and RNA interference.
