Development of Cy5 cyanine dyes for biomedical applications

Abstract:

Diplomová práce se zabývá návrhem a syntézou cyaninových barviv typu Cy5 na bázi heterocyklů 2-methylbenzothiazolu a 2-methylbenzoxazolu. V teoretické části práce je věnována pozornost klasifikaci cyaninových barviv dle délky polymethinového řetězce a charakteru koncových skupin, jejich fyzikálně-chemickým vlastnostem, syntetickým strategiím a biomedicínským aplikacím, zejména s důrazem na fluorescenční sondy využitelné v biologických aplikacích. Experimentální část se soustředí na vícekrokovou syntézu cílových struktur zahrnující modifikaci aromatického jádra, N-alkylaci, tvorbu pentamethinového řetězce a následné funkční úpravy včetně navázání trifenylfosfinu. Připravené intermediáty i cílové produkty byly charakterizovány pomocí NMR spektrometrie, hmotnostní spektrometrie a chromatografických metod. Součástí práce byla rovněž optimalizace reakčních podmínek a separačních technik, včetně sloupcové chromatografie a flash chromatografie. Výsledky ukazují, že benzothiazolové deriváty vykazují příznivější reaktivitu v klíčových syntetických krocích oproti benzoxazolovým derivátům, u nichž se nepodařilo dosáhnout tvorby cílového pentamethinového řetězce. Práce přispívá k rozšíření poznatků o syntéze cyaninových barviv a jejich potenciálním využití jako fluorescenčních sond v biologických aplikacích.

Abstract:

This thesis deals with the design and synthesis of Cy5-type cyanine dyes based on the heterocycles 2-methylbenzothiazole and 2-methylbenzoxazole. The theoretical part of the thesis is focused on the classification of cyanine dyes according to the length of the polymethine chain and the nature of the terminal groups, their physico-chemical properties, synthetic strategies, and biomedical applications, with particular emphasis on fluorescent probes usable in biological systems. The experimental part is focused on the multi-step synthesis of target structures, including modification of the aromatic ring, N-alkylation, formation of the pentamethine chain, and subsequent functionalization, including the attachment of triphenylphosphine. The prepared intermediates and target products were characterized by using NMR spectroscopy, mass spectrometry, and chromatographic methods. The work also includes the optimalization of reaction conditions and separation techniques, including column chromatography and preparative HPLC. The results show that benzothiazole derivatives exhibit more favourable reactivity in key synthetic steps compared to benzoxazole derivatives, where the formation of the target pentamethine chain could not be achieved. This work contributes to expanding knowledge regarding the synthesis of cyanine dyes and their potential usage as fluorescent probes in biological applications.
