Evaluating the Function of Cultured Gastric Cancer Cells

Abstract:

Diplomová práce se zabývá funkční a morfologickou charakterizací a vzájemným porovnáním komerční buněčné linie karcinomu žaludku s primárními buněčnými liniemi nenádorového a nádorového původu odvozenými ze žaludeční tkáně. Teoretická část se věnuje popisu, klasifikaci, epidemiologii a diagnostice karcinomu žaludku. Pozornost je věnována také vybraným buněčným liniím ustanoveným z buněk karcinomu žaludku. Experimentální část je zaměřena na zavedení a optimalizaci stanovení specifických buněčných markerů cytokeratinu 18 a karcinoembryonálního antigenu. Dále se zaměřuje na testování toxicity farmakologicky aktivních látek po inkubaci všech tří linií buněk s kamptothecinem, staurosporinem, cisplatinou a acetaminofenem v časovém intervalu 24 hod. Pro charakterizaci toxického působení vybraných látek jsme stanovovali intracelulární koncentraci glutathionu a dehydrogenázovou aktivitu buněk. Mikroskopickými technikami jsme sledovali také vliv modelových látek na buněčnou morfologii. V poslední části práce jsou prezentovány a diskutovány výsledky.

Abstract:

This thesis deals with the functional and morphological characterization and comparison of the commercially available gastric cancer cell line with primary cell lines of non-tumor and tumor origin derived from human gastric tissue. The theoretical part deals with the description, classification, epidemiology, and diagnosis of gastric cancer. Attention is also paid to selected cell lines established from gastric cancer. The experimental part focuses on the introduction and optimization of assays for detection of specific cell markers cytokeratin 18 and carcinoembryonic antigen. It also focuses on testing the toxicity of pharmacologically active compounds after incubation of all three cell lines with camptothecin, staurosporine, cisplatin and acetaminophen for 24 h. To characterize the toxic effect of the selected compounds, we determined the intracellular glutathione concentration and dehydrogenase activity of the cells. The effect of tested substances was also observed using microscopic techniques. In the following parts of the thesis the results are presented and discussed.
