Abstract:

Cílem této práce je návrh a optimalizace metody úpravy plasmy před MALDI-MS profilováním, které má umožnit včasnou a účinnou detekci nemocí, v tomto případě rakoviny pankreatu, díky rozdílům v profilu plasmy pacienta a kontroly. K dosažení tohoto cíle byly použity metody jako molekulární filtry, deplece abundantních proteinů plasmy, různé afinitní chromatografie, digesce plasmatických proteinů, etc. a jejich kombinace. Dále se na vybrané skupině kontrol a pacientů testovaly kandidátní metody přípravy plasmy, které byly považovány za schopné odlišit profil plasmy pacienta a kontroly. Specifita případného nalezeného biomarkeru je klíčová, neboť některé biomarkery mohou indikovat více onemocnění, nebo například i obyčejnou infekci.

Abstract:

The purpose of this work is to find a suitable procedure of plasma preparation for a MALDI-MS profiling experiment that would enable a cost efficient and early detection of biomarkers for a specific disease, in this case pancreatic cancer. Various plasma preparation techniques such as molecular cut-offs, depletion of most abundant proteins, protein purification kits, proteolytic digestion, etc. have been used in the search for the best preparation technique enabling to distinguish between patient and control. In multiple samples of pacients and controls candidate methods were applied that should deliver differential MS plasma profiles that indicate the presence of pancreatic cancer or should be able to deny it. Specificity of the possible marker is of key importance since some might be matched with multiple diseases or indicate only a nonspecific, common inflammation.
