Malondialdehyd - marker of lipid peroxidation

Abstract:

Malondialdehyd se přirozeně vyskytuje v rostlinných i živočišných buňkách jako vedlejší produkt převážně fotosyntetických procesů. V buňce vystavené stresu jeho koncentrace prudce stoupá v důsledku procesu peroxidace membránových lipidů, který je způsoben reaktivními formami kyslíku či enzymy lipoxygenasami. Malondialdehyd v buňkách aktivuje expresi obranných genů, ale převažují jeho negativní účinky. Je mu připisován vliv na propuknutí Parkinsonovy a Alzheimerovy choroby, genové mutace či stárnutí lidského organismu. U rostlin přímo usmrcuje útočící patogeny, ale poškozuje i buněčné struktury samotné rostliny, což může vést k nekróze napadeného rostlinného pletiva. Cílem praktické části bylo porovnat schopnost elicitinu kryptogeinu a jeho mutantních forem vyvolat peroxidaci lipidů na listech Nicotiana tabacum. Jako spolehlivý marker peroxidace lipidů byl zvolen právě malondialdehyd, který představuje hlavní sekundární metabolit tohoto procesu. K jeho stanovení byla použita metoda tvorby sloučeniny MDA(TBA)2 , která byla detekována pomocí HPLC s reverzní fází. Byl použit rekombinantní kryptogein X24 a jeho mutantní formy – proteiny 56 a L19R. Výsledky ukázaly na schopnost proteinu 56 vyvolat nekrózu pletiva, která je srovnatelná s účinky kryptogeinu, a to i přesto, že 56 neváže mastné kyseliny ani steroly. Protein L19R měl překvapivě stejné účinky jako kryptogein i přes jeho schopnost vázat steroly zhruba s poloviční afinitou než kryptogein.

Abstract:

Malondialdehyde occurs naturally in plant and animal cells as a byproduct of photosynthetic processes. Its concentration sharply increases in cells exposed to stress as a result of membrane lipid peroxidation process, that is caused by reactive oxygen species and enzymes - lipoxygenases . Malondialdehyde activates the expression of defense genes in cells, but its negative features prevail. The impact on the outbreak of Parkinson's and Alzheimer's disease, gene mutations or ageing of human body has been attributed to malondialdehyde. Malondialdehyde directly kills invading pathogens, but also damages the cell structure of the plant itself, that can lead to necrosis of affected plant tissue. The aim of this thesis was to compare the practical ability of elicitin cryptogein and its mutant forms to induce lipid peroxidation in leaves of Nicotiana tabacum. Malondialdehyde was elected as a reliable marker of lipid peroxidation, that is the main secondary metabolite of this process. It was used the method of production MDA(TBA)2 compound for determination of malondialdehyde levels. MDA(TBA)2 was detected by HPLC with reverse phase. It was used the recombinant cryptogein X24 and its mutant forms - 56 and L19R proteins. The results have shown the ability of protein 56 to cause the necrosis of plant tissue, that is comparable with effect of cryptogein in spite of fact that protein 56 does not bind fatty acids or sterols. Protein L19R had surprisingly the same effects as cryptogein despite his ability to bind sterols with half affinity than cryptogein.
