Abstract:

Bialelické nebo též homozygotní delece lze jednoduše definovat jako ztrátu obou alel jednoho genu v genomu buňky. Tyto genetické změny hrají významnou roli při tumorigenezi a progresi celé řady rakovinných onemocnění, neboť často postiženými geny bývají právě důležité tumor supresorové geny. Cílem této diplomové práce je analyzovat soubor dat získaných technikou aCGH s využitím oligonukleotidových DNA čipů firmy Agilent formátu 4x44K u souboru pacientů s mnohočetným myelomem (MM). Práce se soustřeďuje na podrobnou celogenomovou analýzu zisků a ztrát DNA sekvencí a také na charakteristiku nejčastějších nebalancovaných chromozomových aberací. Dalším cílem práce je lokalizovat bialelické delece v genomu MM a zároveň v takovýchto genomových oblastech vybrat vhodné kandidátní geny, které mohou souviset s patogenezí MM, a pomocí RT-PCR tyto delece potvrdit. Práce přispívá k hlubšímu porozumění patogeneze MM a na základě zhotovených celogenomových profilů má umožnit podrobnější stratifikaci pacientů na jednotlivé genetické podskupiny, které by se vyznačovaly odlišnou prognózou.

Abstract:

Biallelic or homozygous deletions can be simply defined as the loss of both alleles of a gene in the genome of the cell. These genetic changes play an important role in tumorigenesis and progression of a great variety of cancers, because the affected genes are very often important tumor-suppressors. The aim of this thesis is to analyze the data set obtained by aCGH technology using oligonucleotide microarrays Agilent format 4x44K in patients with multiple myeloma (MM). The work focuses on a detailed analysis of whole genome gains and losses of DNA sequences and the characteristics of the most common unbalanced chromosome aberrations in the genome of tumor cells. Another goal is the localization of biallelic deletions in the genome of MM, and in such genomic loci select candidate genes that may be related to the pathogenesis of this disease, and by RT-PCR confirm these deletions. The work contributes to a deeper understanding of the pathogenesis of MM and should allow further stratification of patients, based on genome-wide profiles, into different genetic subgroups which will be characterized by different prognosis.
