The role of antioxidants in the prediction of honey bee, Apis mellifera L., longevity

Abstract:

Antioxidanty slouží jako ochrana proti vzniku oxidativního stresu, který může být pro organismus fatální. Degenerace organismu vlivem oxidativního stresu je součástí mnoha teorií stárnutí, a proto mohou být množství a aktivita antioxidantů zásadní při predikci dlouhověkosti. Včela medonosná (Apis mellifera) je vhodným modelem pro zkoumání vlivu antioxidantů na délku života, jelikož se v našem podnebném pásmu vyvíjí dvě odlišné populace včel: krátkověké letní a dlouhověké zimní včely. Krátkověké včely žijí několik týdnů, dlouhověké se však mohou dožívat až devíti měsíců. V rámci studia sezónních změn v aktivitě a koncentraci antioxidantů v hemolymfě včelích dělnic bylo zjištěno, že ačkoli se během roku nemění celková antioxidační kapacita, hladiny dílčích antioxidantů kolísají. Koncentrace antioxidantu glutathionu v hemolymfě pozitivně koreluje s teplotou a vyskytuje se u včel převážně v letních měsících. Podle předběžných dat mají včely v hemolymfě na začátku léta také vysoké množství vitaminu C. Oproti tomu množství proteinu vitellogeninu s antioxidačními schopnostmi dominuje v hemolymfě během zimních měsíců a negativně koreluje s teplotou. Podmínky dané geografickou polohou včelnice, kterými může být např. rozdílná potravní nabídka či počasí, ovlivňují celkové množství antioxidantů, glutathionu i vitellogeninu v hemolymfě včel. Na všech včelnicích byly pozorovány stejné rozdíly mezi letními a zimními včelami, ale při srovnání jednotlivých včelstev v určité části sezóny mezi sebou byly hladiny zmíněných antioxidantů různé. Stresové faktory jako parazitace a transport včel antioxidační parametry ovlivňují jen minimálně. Pro predikci dlouhověkosti včel tedy není vhodným znakem celková antioxidační kapacita, dlouhověké zimní včely se však od letních odlišují vysokou koncentrací vitellogeninu a nižším množstvím glutathionu v hemolymfě.

Abstract:

Antioxidants are the defence system against the harmful effect of oxidative stress. Oxidative stress can be fatal for an organism. Degradation of the organism in connection with the oxidative stress is included in many theories of senescence. That is the reason for choosing a concentration of antioxidants as a marker for the prediction of longevity. Honey bee (Apis mellifera L.) is a suitable experimental model for studies focused on effect of antioxidants on longevity, because there are two different population of honey bees in the temperate climate: short-living summer bees and long-living winter bees. Short-living bees live for a few weeks; however, long-living bees can survive even nine months. It was found out that individual antioxidative components of the hemolymph vary, although the total antioxidant capacity of bee worker’s hemolymph remains stable during the year. The concentration of antioxidant glutathione in the hemolymph correlates positively with the temperature and it is present in higher levels in the hemolymph of bees throughout the summer season. Vitamin C with antioxidative abilities appears to be abundant in the hemolymph during the early summer according to the preliminary data. In contrast to these two antioxidants, the antioxidative protein vitellogenin dominates the hemolymph in the winter season. Total antioxidant capacity, vitellogenin and glutathione levels in hemolymph depend on geographic location of apiaries, where the bees were collected, presumably because they differ in food sources, weather conditions etc. Differences between summer and winter bees are the same in all tested apiaries as observed in the experiment, which followed seasonal changes in antioxidants, but there were variations within individual bee colonies. Although, infestation and transport could be stressful for honey bees, it has nearly no effect on the antioxidative parameters. The main conclusion of the thesis is that the total antioxidant capacity is not a suitable parameter for the prediction of honey bee longevity; however, the long-living honey bees could be distinguished from the short-living ones based on their higher level of vitellogenin and the lower amount of glutathione in the hemolymph.
