Mutation motifs in DNA sequences

Abstract:

DNA je nositelkou genetické informace všech živých organismů a některých virů. U těchto organizmů umožňuje přenos dědičné informace do dalších generací. Mutace v DNA jsou součástí mechanismu, který organismům umožňuje přizpůsobit se změnám ve vnějších podmínkách. Mohou ale také způsobovat dědičná onemocnění a rakovinu. Vznik mutací je ovlivněn především přesností replikace DNA, činností opravných mechanismů a mírou vystavení mutagenním faktorům. Frekvence výskytu mutací se v rámci nukleotidové sekvence liší. Pozice, na kterých dochází k mutacím velmi často, se označují jako hotspoty. Pozice se vzácným výskytem mutací se označují jako coldspoty. Znalost těchto motivů je důležitá pro pochopení mechanismu vzniku mutací a fungování opravných mechanismů. Tato práce je zaměřena na popis typů mutací v sekvencích DNA, mechanismy jejich vzniku a oprav a také na známé mutační motivy.

Abstract:

DNA is a carrier of the genetic information of all living organisms and some viruses. In these organisms DNA allows transfer of the genetic information into next generations. Mutations in DNA are a part of the mechanism that enables organisms to adapt to the changes in the environment. Howewer, mutations may also cause inherited diseases and cancer. Emergence of mutations is influenced mainly by DNA replication accuracy, repair mechanisms activity and exposure to mutagenic factors. The frequency of mutations within a nucleotide sequence differs. Positions at which mutations occur very often are referred to as hotspots. Position with rare mutation occurrence are referred to as coldspots. Knowledge of these motifs is important for understanding the mechanism of the emergence of mutations and the mechanism of mutation repair. This thesis focuses on description of the mutation types in DNA sequences, emergence of mutations, repair mechanisms, and also on known mutation motifs.
