Ambient air contamination - identification of toxic potential of pollutants and their mixtures

Abstract:

V této bakalářské práci se zabývám problematikou znečištění vnějšího ovzduší a jeho vlivu na zdraví člověka. Zaměřuji se především na kyslíkaté deriváty polycyklických aromatických uhlovodíků, které se mohou v ovzduší vyskytovat ve významných koncentracích, ale je velmi málo informací ohledně jejich možného příspěvku k toxickému potenciálu směsí látek v ovzduší. V první části práce shrnuji především fyzikálně-chemické vlastnosti a koncentrace těchto látek v ovzduší, které bývají mnohdy srovnatelné s koncentrací parentálních polycyklických aromatických uhlovodíků. V další části se pak zabývám jejich potenciálem k biologickým účinkům, zejména k endokrinní disrupci, který je často stejně významný jako u jejich parentálních sloučenin, ale není řádně prozkoumán. U řady vzorků ovzduší bylo potvrzeno působení na estrogenní, androgenní či arylhydrokarbonový receptor ve kterém by kyslíkaté deriváty polycyklických aromatických sloučenin mohly hrát významný podíl. Praktická část této práce přispívá k hodnocení potenciálů relevantních kyslíkatých derivátů polycyklických aromatických uhlovodíků působit těmito mechanismy, což umožní charakterizaci jejich podílu na účincích vzorků z prostředí. V praktické části jsem otestovala estrogenním a arylhydrokarbonovým receptorem zprostředkovanou aktivitu čtyř látek, jejichž přítomnost v ovzduší byla zjištěna na Ostravsku výzkumem ústavu RECETOX. Estrogenní účinky vykazovaly látky benzantron, 1,4-antrachinon a 5,12-naftacenchinon. 9,10-fenantrenchinon estrogenní aktivitu neindukoval. Aktivitu arylhydrokarbonového receptoru pak indukovaly látky 1,4-antrachinon a 5,12-naftacenchinon. Látky benzantron a 9,10-fenantrenchinon aktivitu nevyvolaly. V této práci tak potvrzujeme potenciál kyslíkatých derivátů polycyklických aromatických uhlovodíků hrát roli v endokrinní disrupci. Biologické účinky těchto derivátů je však nutné ještě detailněji prozkoumat.

Abstract:

In this thesis I deal with the issue of pollution of the ambient air and its influence on human health. I focus primarily on oxygen derivatives of polycyclic aromatic hydrocarbons, which can be abundant in the atmosphere, but there is little known regarding their potential contribution to toxic potential of compounds mixtures in the atmosphere. In the first part of the thesis I summarize mainly the physico-chemical properties and the concentration of these compounds in ambient air, which is often comparable with concentration of the parent polycyclic aromatic hydrocarbons. In the next part I deal with their potential for biological effects, especially for endocrine disruption, which are often similarly important as in parental polycyclic hydrocarbons, but there has been no detailed research. For many air samples the effect on the estrogenic, androgenic or arylhydrocarbon receptors has been confirmed and the oxygen derivatives could play a role in it. The practical part of this work contributes to the evaluation of the potentials of the relevant oxygen derivatives of polycyclic aromatic hydrocarbons to act via these mechanisms, which will allow characterization of their possible role in the effects of the environmental samples. In the practical part I tested the estrogenic and arylhydrocarbon receptor-mediated activity of four substances, whose presence in the air was detected in Ostrava during RECETOX research study. Benzanthrone, 1,4-anthraquinone and 5,12-naphthacenequinone were shown to be estrogenic while 9,10-fenantrenchinone did not induce estrogenic activity. The activity of the aryl hydrocarbon receptor was induced by 1,4-anthraquinone and 5,12-naphthacenequinone while benzanthrone and 9,10-phenanthrenequinone did not cause any AhR-mediated activity. In this thesis we confirm the potential of oxygen derivatives of polycyclic aromatic hydrocarbons to play a role in endocrine disruption. However, the biological effects of these compounds need to be explored further.
