Distribution of Sympetrum depressiusculum populations and their habitat requirements in Europe

Abstract:

Vážku rumělkovou, Sympetrum depressiusculum (Selys, 1841) řadíme do rodu Sympetrum. Jedná se o menší vážku, jejímž hlavním poznávacím znakem jsou černé nohy a výrazné zbarvení těla, které je odlišné na základě pohlaví. Samci jsou zbarvení červeně, oproti samicím, které dominují žlutooranžovou barvou. U obou pohlaví se vyskytují charakteristické párové trojúhelníkovitě protáhlé černé skvrny na bocích zadečku. V Evropě je její výskyt zaznamenán pouze lokálně, pokračující na východ až po Japonsko. Z důvodu lokálního výskytu a úbytku jejího počtu ve srovnání s historickými údaji, řadíme tento druh mezi kriticky ohrožený. Tato práce se dále zabývá habitatovými nároky a distribucí této vážky. Na základě zjištěných informací z dostupné literatury, byly sestaveny mapy Evropy a České republiky, které graficky zaznamenávají výskyt druhu S. depressiusculum. V rámci praktické části byla provedena základní genetická analýza celkem čtyř populací druhu S. depressiusculum z různých států, s cílem zjistit genetickou příbuznost zmíněných populací a vybrat kandidátní genetické markery o dostatečné variabilitě pro složitější analýzy. Využité markery cytochrom c oxidáza 1, cytochrom c oxidáza 2 a elongační faktor 1-?, nevykazovaly dostatečnou variabilitu, tudíž je potřeba ve výzkumu dále pokračovat a najít variabilnější genetické markery vhodné pro studium příbuznosti evropských populací.

Abstract:

Spotted Darter, Sympetrum depressiusculum (Selys, 1841) belongs to the genus Sympetrum. It is a smaller dragonfly, whose main identification characters are black legs and a distinctive body color, which is different based on sexes. Males are red, in contrast to females, who are mainly yellow-orange. In both sexes, there are characteristic paired triangular elongated black spots on the sides of the abdomen. In Europe, its occurrence is recorded only locally, continuing east to Japan. Due to the local occurrence and decrease in its number in comparison with historical data, we classify this species as critically endangered. This work further deals with the habitat requirements and distribution of this dragonfly. Based on the available literature sources, maps of Europe and the Czech Republic were created, which graphically record the occurrence of S. depressiusculum. The practical part of the work deals with basic genetic analysis of four populations of S. depressiusculum from different countries, in order to determine the genetic relationship of these populations and select candidate genetic markers of sufficient variability for more complex analyzes. The used markers cytochrome c oxidase 1, cytochrome c oxidase 2 and elongation factor 1? did not show sufficient variability, therefore it is necessary to continue the research and find more variable genetic markers suitable for studying the relationships of European populations.
