Characterisation of Flavivirus neutralisation by antibodies using cryo-electron microscopy

Abstract:

Flaviviry jsou rod obalených virů s jednořetězcovou RNA patřících do čeledi Flaviviridae přenášených členovci. Do rodu patří i virus klíšťové encefalitidy (TBEV), který je jedním z flavivirů přenášených klíšťaty. Infekce TBEV jsou endemické v Asii a Evropě, přičemž závažnost onemocnění se liší podle kmene viru. Přestože je dostupné očkování, počet případů každoročně stoupá. Klíšťová encefalitida postihuje centrální nervový systém, kde vyvolává zánět, který může být smrtelný. V současnosti neexistuje žádná cílená léčba, a tak se lékaři musí spoléhat pouze na podpůrnou péči. Zde by mohly být užitečné protilátky – bylo totiž prokázáno, že jejich podání má neuroprotektivní účinky, a vede k méně závažnému průběhu onemocnění u myší. Nicméně poznatky o molekulárních mechanismech neutralizace TBEV protilátkami jsou omezené. Za využití virových částic vyizolovaných z infikovaných tkáňových kultur, kryo-elektronové mikroskopie a „single particle“ analýzy jsme vyřešili struktury zralých částic TBEV z kmenů Neudörfl a Hypr s vyšším rozlišením než jakékoli dříve publikované struktury. Dále jsme vyřešili struktury dvou komplexů TBEV s Fab fragmenty, A4 a IC3, specifickými pro dvě odlišné domény obalového proteinu TBEV. Prokázali jsme, že vazba těchto Fab fragmentů narušuje organizaci virového obalu, a tak může neutralizovat virus tím, že brání jeho navázání na buněčné receptory. Pochopení různých mechanismů neutralizace flavivirů může v budoucnu pomoci s vývojem terapeutických protilátek pro léčbu infekcí TBEV.

Abstract:

Flaviviruses are a genus of single-stranded positive-sense RNA viruses belonging to the family Flaviviridae. Arthropod vectors are the main mode of transmission for the viruses in the genus. The tick-borne encephalitis virus (TBEV) is the causative agent for tick-borne encephalitis and one of the Flaviviruses is transmitted by ticks. TBEV infections are endemic to Asia and Europe with the severity of the disease varying by virus strain. Although vaccination is available, the number of cases is increasing annually. Tick-borne encephalitis affects the central nervous system where it establishes an inflammation, which may be fatal. Nevertheless, there are currently no targeted treatment options, and thus doctors rely solely on supportive care. This is where antibodies may be of use – as it was demonstrated that their admission has neuroprotective effects and hence leads to a less severe course of disease in mice. However, insights into the molecular mechanisms of TBEV neutralisation are limited. Using viral particles purified from infected tissue culture cells, cryo-electron microscopy, and single particle analysis, we solved the structures of mature particles of TBEV from strains Neudörfl and Hypr to a higher resolution than any previously published structures. Furthermore, we have solved structures of complexes of TBEV with two Fab fragments, A4 and IC3, specific for two distinct domains of the TBEV envelope protein. We have demonstrated that binding of these Fab fragments disrupts the organisation of the viral envelope and thus may neutralise the virus by preventing its attachment to cellular receptors. Understanding various mechanisms of flavivirus neutralisation might aid with tailoring therapeutic antibodies to treat TBEV infections in the future.
