Uptake and translocation of fluoranthene in pea plants

Abstract:

Cílem diplomové práce bylo hodnocení kořenového příjmu polycyklického aromatického uhlovodíku (PAH) fluorantenu (FLT) a jeho následné translokace a distribuce po 2, 4, 6, 8, 24 a 30 hodinách u 10-denních rostlin hrachu setého (Pisum sativum L.), které byly kultivovány v Reid-Yorkově živném roztoku s přídavkem směsi fluorantenu (0,1 mg/l) a 14C-FLT (5 µl/50 ml, o celkové aktivitě 1110000 dpm/0,5 µCi). Dále byl hodnocen i foliární příjem fluorantenu a jeho následná translokace a distribuce po 2, 4, 6, 24, 48 a 72 hodinách u rostlin hrachu, které byly kultivovány v Reid-Yorkově živném roztoku a u kterých byl na druhý, plně vyvinutý list nanesen 14C-FLT v kapce destilované vody (1µl 14C-FLT v 10 µl destilované vody). Využitím metody radioaktivního izotopu uhlíku 14C bylo prokázáno, že rostliny hrachu jsou schopné přijímat fluoranten jak kořenovým systémem, tak foliární cestou a během krátké doby (2 hod.) tuto látku nebo produkty její transformace distribuovat do ostatních částí rostliny jak akropetálně, tak i bazipetálně.

Abstract:

The aim of thesis was to assess the root uptake of polycyclic aromatic hydrocarbon (PAH) fluoranthene (FLT), its transport and distribution in 10 days old seedlings of Pisum sativum L. during 2, 4, 6, 8, 24 and 30 hours of exposure period. The plants were cultivated in Reid-York nutrient solution with fluoranthene (0,1 mg/l) and 14C-FLT (5 µl/50 ml, total acitivy 1110000 dpm/0,5 µCi). Also the assessement of foliar uptake of FLT, its transport and distribution in 10 days old seedlings of pea during 2, 4, 6, 24, 48 and 72 hours exposure period was done, when the plants were cultivated in Reid-York nutrient solution and their second fully-developed leaf was contamined by 14C-FLT in a drop of distilled water (1µl 14C-FLT in 10 µl distilled water). The use of radioactive isotope 14C method shows, that pea seedlings were able to take up the FLT by roots and leafs and to ditribute this chemical or products of its transformation during short time (2 hours) into others parts of plants, both acropetally and basipetally.
