Colonization dynamic of human intestinal tract with antibiotic resistant bacteria

Abstract:

Tato diplomová práce se zabývá problematikou antibiotické rezistence bakterií identifikovaných v rektálních výtěrech kojence a jeho matky. Sledována byla dynamika kolonizace trávicího traktu dítěte jednotlivými druhy čeledi Enterobacteriaceae společně s probiotickým kmenem Escherichia coli, který byl dítěti z důvodu průjmového onemocnění podán. Bakteriální izoláty byly identifikovány a charakterizovány pomocí metod molekulární biologie, současně byla otestována schopnost probiotického kmene přijímat geny antibiotické rezistence prostřednictvím konjugace. V souboru vzorků byly identifikovány pouze dva bakteriální druhy, a to Escherichia coli a Klebsiella oxytoca. Byly nalezeny kmeny rezistentní k antibiotikům ve vzorcích od matky i dítěte. Přítomnost probiotika v trávicím traktu dítěte byla pozorována po několik týdnů. Probiotický kmen E. coli přijímal geny rezistence s frekvencí 10-6–10-5. Potvrdilo se tak, že by se mohl stát rezervoárem antibiotické rezistence v trávicím traktu lidí.

Abstract:

In this diploma thesis the problem of antibiotic resistant bacteria found in rectal swabs from an infant and its mother is evaluated. Colonization dynamic of child’s intestinal tract with bacteria belonging to Enterobacteriaceae family as well as with Escherichia coli probiotic strain that was applied to the child because of diarrhoea was monitored. Bacterial isolates were identified and characterized using molecular biology methods, the ability of the probiotic strain to acquire antibiotic resistance genes via conjugation was tested. Only two bacterial species Escherichia coli and Klebsiella oxytoca were found in the samples. Antibiotic resistant strains were detected in the samples from the mother and the child. The presence of the probiotic strain in the child’s intestinal tract was seen for several weeks. The probiotic strain acquired resistance genes with frequency from 10-6 to 10-5. This result confirmed the ability of probiotic strain E. coli to become a reservoir of antibiotic resistance genes in the human intestinal tract.
