Functions of nitric oxide in insect immune system

Abstract:

V teoretické části jsou shrnuty informace týkající se metabolismu, biologických a imunitních funkcí oxidu dusnatého (NO) u hmyzu. Je popsán mechanismus vzniku NO, cíle jeho působení a jeho funkce v nervovém systému hmyzu. Rovněž jsou popsány funkce NO v imunitním systému hmyzu, ve kterém je NO zapojen jako signální a cytotoxická molekula. Poslední část je věnována funkci NO v imunitním systému včely medonosné, modelového organismus v experimentální části. V experimentální části zaměřené na zavedení metod detekce a analýzy enzymu S-nitrosoglutathionreduktasy (GSNOR, EC 1.1.1.284) ve vzorcích tkání včely medonosné byly použity vzorky ze 4 různých včelstev: 3 klinicky zdravých a 1 včelstva kolabujícího na varroózu. Bylo zjištěno, že koncentrace proteinů byla v případě včelstva kolabujícího na varroózu o polovinu nižší než u klinicky zdravých včelstev. Dále byla stanovena aktivita GSNOR a bylo zjištěno, že aktivita GSNOR je napříč včelstvy podobná. GSNOR byla inhibována inhibitorem lidské GSNOR látkou N6022. Imunodetekcí byla potvrzena přítomnost GSNOR ve všech částech včelího těla po předchozí separaci proteinů ze vzorků extraktů metodou SDS-PAGE a Western blot. Nativní elektroforézou a následným barvením s využitím dehydrogenasové aktivity GSNOR byla přítomnost GSNOR potvrzena pouze v hrudi a zadečku včely medonosné.

Abstract:

The theoretical part summarizes information concerning the metabolism, biological and immune functions of nitric oxide (NO) in insects. Mechanisms of NO formation, its targets and its function in the nervous system of insect are also described. NO functions in insect immune system in which NO is involved as a signal and cytotoxic molecule are summarized. The last part deals with the function of NO in the immune system of the honey bee, which is the model organism in the experimental part. In the experimental part, focused on the implementation of methods detection and analysis of S-nitrosoglutathionereductase (GSNOR, EC 1.1.1.284) in samples of honey bee tissues, were used samples from 4 different hives: 3 clinically healthy bee colonies and 1 bee colony collapsing on varroosis. We found, that the concentration of proteins in hives collapsing on varroosis was a half lower than in healthy hives. Furthermore, GSNOR activity was determined and GSNOR aktivity across colonies was found to be similar. GSNOR was inhibited by the human GSNOR inhibitor N6022. Immunodetection confirmed the presence of GSNOR in all parts of the bee body after previous separation of proteins from extracts samples by SDS-PAGE and Western Blot. Native electrophoresis, followed by staining using the dehydrogenase activity of GSNOR, confirmed the presence of GSNOR only in the thorax and abdomen of honey bees.
