Srovnávací topografie telomerových oblastí v lidských buňkách s různými mechanismy prodlužování telomer

Abstract:

Cílem této diplomové práce bylo porovnání lokalizace telomer a porovnání výskytu TA ve zdravých buňkách, v nádorových buňkách s aktivní telomerázou a v nádorových buňkách s ALT mechanismem prodlužování telomer. Tyto parametry byly studovány pomocí subtelomerických DNA sond v rámci opakované dvoubarevné FISH na 2D fixovaných jádrech, dále pomocí HRCM a následné softwarové analýzy. Jako modelový materiál byly zvoleny tyto plicní buněčné linie: WI-38 (zdravé plicní fibroblasty), nádorová linie HT-1080 s aktivní telomerázou a linie WI-38 VA-13/2RA (VA-13) s ALT mechanismem prodlužování telomer, u které existoval předpoklad častějších TA, právě z důvodu těsnější lokalizace při rekombinačním prodlužování telomer. Pro tyto studie byly vybrány chromozomy 2, 4, 8 a 9. Ve většině případů byly distribuce radiálních vzdáleností všech zkoumaných telomer u všech tří typů buněčných linií shodné nebo porovnatelné až na pár výjimek, mezi které patří především výrazný posun polohy obou telomer chromozomu 2 v buňkách HT-1080. Při porovnání poloh obou telomer konkrétních chromozomů ve všech třech typech buněk se v souladu s dřívějšími výsledky nalezla centrálnější poloha telomery q- v porovnání s telomerou p- v případě chromozomu 9 ve zdravých buňkách WI-38 a dále v případě chromozomu 2 ve všech třech typech buněk. Naprosto opačné radiální pozice však vykazují telomery chromozomu 4 ve všech třech typech buněk a také telomery chromozomu 8 v buňkách WI-38. Na druhou stranu, radiální distribuce obou telomer chromozomů 8 a 9 v buňkách VA-13 a pro chromozom 9 v buňkách HT-1080 jsou v podstatě totožné. U všech telomer chromozomů 8 a 9 byl potvrzen předpoklad o vyšším podílu telomerických asociací u buněk s ALT mechanismem prodlužování telomer oproti buňkám s aktivní telomerázou nebo zdravým buňkám. Obdobně systematický trend se však nepotvrdil u telomer chromozomů 2 a 4, kde nejvíce telomerických asociací bylo nalezeno u telomer buněk HT-1080 s aktivní telomerázou.

Abstract:

The comparative topography of telomeres in human cells with different lengthening mechanism Main goal of this work was to compare the telomeric localizations and number of occurrences of TAs in healthy cells, in tumor cells with active telomerase, and in tumor cells with ALT mechanism. The objectives were pursued by visualizing telomeres with subtelomeric DNA probes by repeated double-color FISH on 2D fixed nuclei, observed using HRCM and followed by software analysis of acquired data. As models were chosen following lung tissue cell lines: cell line WI-38 (healthy lung fibroblasts), tumor cell line HT-1080 with active telomerase, and cell line WI-38 VA-13/2RA (VA-13) with ALT mechanism, which was expected to have higher occurrence of telomere associations due to tight localization of telomeres during recombinatory lengthening of telomeres. We have chosen telomeres of chromosomes 2, 4, 8 and 9 for these studies. Distributions of radial distances of all analyzed telomeres for all assessed cell lines are comparable in most cases with only several exceptions. Among these exceptions belongs significant shift of localization of both telomeres of chromosome 2 in HT-1080 cell line. The comparison of localization of both telomeres of the chromosomes in all three cell lines is in agreement with previous results, where we have found the localization of telomere q- more central in comparison to telomere p- for chromozome 9 in healthy WI-38 cells, and for chromozome 2 in all three cell lines. Rather opposite results were obtained for telomeres of chromozome 4 in all three cell lines and for telomeres of chromozome 8 in WI-38 cell line. On the other hand, radial distributions of both telomeres of chromosomes 8 and 9 in VA-13 cell line and chromozome 9 in HT-1080 cell line are practically equal. Both telomeres of chromosomes 8 and 9 have higher occurrence of telomere associations in ALT cell line with alternative telomere lengthening in comparison to cells with active telomerase. Similarly systematic tendency was not found for telomeres of chromosomes 2 and 4, which poses the highest occurrence of telomeric associations in HT-1080 cell line; cell line with active telomerase.
