Preclinical evaluation of enzybiotic components designed by the in-silico method for creating mutants with the desired effect.

Abstract:

Infekce způsobované rezistentními patogenními bakteriemi jsou v součastnosti jedny z nejvážnějších problémů nejen v klinickém prostředí ale i v lidské populaci po celém světe. Jednou z těchto bakterií je Staphylococcus aureus. Díky své vynikající schopnosti adaptace na nepříznivé podmínky se mnohé kmeny této bakterie staly rezistentními k téměř všem v současné době používaným antibiotikům. Jako alternativní léčba takovýchto infekcí byly už v minulosti používané pro jejich bezpečnost a efektivitu lytické enzymy bakteriofágů. Navzdory tomu jsou mnohé z nich z důvodu jejich nízké stability reálně nepoužitelné. S rozvojem metod proteinového inženýrství a lepšímu porozumění faktorů přispívajících k této stabilitě se tenhle problém dá vyřešit. Cílem praktické části této bakalářské práce je připravit mutantní variantu endolysinu z bakteriofága 812 se zvýšenou stabilitou při zachování funkčnosti tohoto proteinu. Teoretická část přináší základní znalosti jak o lytických enzymech působících na Staphylococcus aureus, tak i o metodách proteinového inženýrství používaných k účelům zvýšení stability enzymů.

Abstract:

Infections caused by resistant pathogenic bacteria are currently one of the most severe problems, not only in clinical conditions, but in communities throughout the world as well. One of these pathogenic resistant bacteria is Staphylococcus aureus. Thanks to its remarkable adaptability to unfavourable conditions many strains of this bacterium have become resistant to almost all presently used antibiotics. Bacteriophage lytic enzymes have already been used as an alternative treatment for these infections in the past, mostly because of their safety and effectiveness. Despite that, many of them are not realistically usable as a result of their low stability. However, due to constantly developing protein engineering methods and a better understanding of the factors enhancing this attribute, the stability problem is solvable. The main objective of this work is, therefore, to prepare a mutant variant of the endolysin from bacteriophage 812 with an increased stability, while retaining its original activity. The theoretical background of this work presents basic knowledge about Staphylococcus aureus, lytic enzymes and also protein engineering methods used for the purpose of increasing enzyme stability.
